As an important food,functional food is more and more favored and valued by people.Poor gastrointestinal function and poor digestive function is an important factor that puzzles public health.Therefore,the research and development of functional food that can improve gastrointestinal function and promote digestion has great social value and market application prospect.Based on the traditional Chinese medicine formula for regulating the spleen and stomach,this study developed a food to improve gastrointestinal function.The relevant research contents and results are as follows:(1)Optimization of the formula of Polygonatum and Euryale ferox powder based on the formula theory of traditional Chinese Medicine:Guided by the TCM theory of Invigorating Qi,invigorating spleen and nourishing blood,this study,after consulting a large number of relevant TCM research materials and formula comparisons,and consulting relevant TCM experts,combined with the current"medicine and food homologous"list of traditional Chinese medicine,replaced the non medicine and food homologous traditional Chinese Medicine with the medicine and food of the same flavor on the basis of the traditional Chinese medicine polygonatum Euryale decoction.The dietotherapy formula for obtaining Polygonatum and Euryale ferox powder is 1 polygonatum,2Euryale,1 yam,1 wild jujube kernel,1 jujube,2 Codonopsis pilosula and 0.5patchouli.With the research method of modern traditional Chinese medicine network pharmacology,the related pharmacological effects of dietotherapy formula of Polygonatum and Euryale ferox powder were studied theoretically.The results showed that active ingredients in the formula could affect the content of reactive oxygen species,improve oxidative stress and prevent cells from oxidative damage by regulating STAT3,SRC,PIK3R1 and other target proteins;It contains a variety of natural antioxidants,such as sinomenine,aconitine,diosgenin,etc,which can inhibit the decline of antioxidant enzyme level and digestive enzyme activity in the body,and significantly improve functional dyspepsia.(2)Evaluation of synergistic antioxidation of Polygonatum and Euryale ferox powder:This experiment was conducted to study the synergistic antioxidation between the raw materials of Polygonatum and Euryale ferox powder and the effects of different production processes on the antioxidation of Polygonatum and Euryale ferox powder.The results show that the IC50 values of DPPH and ABTS were 11.89mg·m L-1and 5.89 mg·m L-1,and the IC50 values of direct steaming were 14.18mg·m L-1and 7.78 mg·m L-1.The preparation of Polygonatum and Euryale ferox powder by soaking and steaming has stronger antioxidant activity.Finally,the soaking and steaming method was selected as the research method of the follow-up process.Through direct method and algebraic addition,the synergistic antioxidation of various raw materials of Polygonatum and Euryale ferox powder was compared.When the concentration was 32 mg·ml-1,the addition and clearance rate of each single raw material component was 84.52%,which was less than the actual measured value of87.45±0.81%,indicating that the ingredients in the formula had synergistic antioxidation.(3)Optimization of preparation process of Polygonatum and Euryale ferox powder by Response Surface Methodology:In this experiment,the soaking steaming method was used as the preparation process,and the content of total flavonoids and total polysaccharides was used as the evaluation index.Through response surface design,the optimal steaming process conditions were determined as 1:29(raw material:soaking water),soaking time 0.5 h and steaming time 0.9 h.The actual measured values of total flavonoids and total polysaccharides were 8.43±0.11 mg RE·g-1and 38.60±0.63%respectively.With the increase of the particle size of raw materials,the dispersity of Polygonatum and Euryale ferox powder gradually decreases,the solubility first increases and then decreases,and it is easy to condense into blocks.Finally,80 mesh is the best particle size of Polygonatum and Euryale ferox powder.Through Plackett-Burman method,it was determined that erythritol,skimmed milk powder and konjac powder had a significant impact on sensory evaluation,and through response surface design,the optimal formula conditions were determined as follows:Polygonatum and Euryale ferox powder accounted for 77.61%,erythritol accounted for 1.94%,skimmed milk powder accounted for 10.86%,konjac powder accounted for 1.05%,chrysanthemum powder accounted for 7.76%,stevia glycoside accounted for 0.78%,and the sensory score of this formula was 85.71.(4)Effects of Polygonatum and Euryale ferox powder on fatigue,antioxidation and intestinal function in mice with functional dyspepsia:In this experiment,a mouse model of functional dyspepsia was established by compound factor method to study the effects of Polygonatum and euryale seed powder on the symptoms and signs(body weight and diet),fatigue resistance,spleen and thymus index,serum antioxidant enzyme activity,serum digestive enzyme activity and intestinal flora of mice.The results showed that the mice treated with wine vinegar rhubarb method showed symptoms of mental fatigue,lethargy and laziness,yellow hair,dirty ears,soft stool,decreased diet and weight.The symptoms of the model group lasted until the 28 days,indicating that the model was successfully established.Compared with the model group,the middle and high dose groups could significantly improve the symptoms and signs of mice,and the stay time of rotating rod and swimming time were significantly increased(P<0.05);The spleen index and thymus index of mice in middle and high dose groups increased significantly(P<0.05).Polygonatum and Euryale powder could improve the activities of antioxidant enzymes and digestive enzymes in the serum of mice.In the high dose group,MDA was significantly decreased(P<0.01),SOD and GSH-PX were significantly increased(P<0.01);The amylase activity was significantly increased in the middle and high dose groups(P<0.05),while the lipase activity was significantly increased only in the high dose group(P<0.05).The middle and high dose groups significantly increased the number of bifidobacteria and lactic acid bacteria in the intestinal tract of mice(P<0.01),and significantly decreased the number of Escherichia coli and Enterococcus(P<0.05).The results showed that Polygonatum and Euryale ferox powder could improve the physical signs,exercise and fatigue resistance,improve the immunity of the body,protect the body from free radical attack,promote digestion and absorption,regulate the number of intestinal microorganisms,and then improve the functional dyspepsia.
