Hemerocallis Fulva flower is the flower bud of the Hemerocallis Fulva.It is rich in phenolic compounds and has anti-oxidant,anti-cancer and anti-depressant effects.At present,there are few studies on the antioxidant effect of phenolic compounds in Hemerocallis Fulva flowers and their efficacy evaluation in cosmetics.Based on this,this study used five colors of Hemerocallis Fulva flowers as raw materials,and under three different processing methods,the variety with better antioxidant properties were obtained by comparing the content of total flavonoids and total polyphenols and the scavenging rate of free radicals;To determine the best form of phenolic compounds,the three forms of phenolic compounds extracted from it were measured by HPLC to explore the relationship between the composition and content of phenolic compounds and the antioxidant activity in Hemerocallis Fulva flowers;Finally,It is applied to cosmetic emulsions to evaluate its antioxidant effect combined with cell experiments.The main research conclusions are as follows:(1)As the color of the Hemerocallis Fulva flowers deepen,the content of total flavonoids and total polyphenols in the Hemerocallis Fulva flower increases.After enzymolysis-ultrasound and fermentation-ultrasound treatment,the content of total flavonoids and total polyphenols increased.As the comparison between the two,fermentation-ultrasound had the best effect,and the content of total flavonoids and total polyphenols of the red parrot after fermentation-ultrasound reached the highest,the content reached 57.5mg/g and34.1mg/g respectively,and had an increase of 10.58%and 13.67%compared with that before fermentation;Under the fermentation-ultrasonic treatment,the red parrot had the highest removal efficiency of DPPH and ABTS,and the IC50values of them reached 44.60?g/m L and26.72?g/m L respectively;The correlation analysis shows that the content of total flavonoids and total polyphenols and DPPH and ABTS removal rate has a good correlation,which proves that the phenolic compounds in Hemerocallis Fulva flowers have an important contribution to the antioxidant activity.(2)Among the three phenolic components of the red parrot,the content of free total flavonoids and total polyphenols is the highest,accounting for more than 75%of the total content;after enzymolysis and fermentation,the content of free flavonoids and polyphenols increased significantly(p<0.05),the content of bound flavonoids and polyphenols decreased significantly(p<0.05).Fermentation-ultrasonic extraction of free phenol has the best effect on DPPH and ABTS clearance,and the IC50of them values reached 40.53?g/m L and22.10?g/m L respectively.Through the analysis of HPLC,both enzymatic hydrolysis and fermentation significantly increased the content of various free phenols in the red parrot,and newly detected p-coumaric acid,and content of free quercetin tripled after fermentation;The correlation analysis showed that chlorogenic acid,rutin,caffeic acid and quercetin are highly correlated with antioxidant activity,and has a greater contribution to antioxidant activity in the free state.(3)The free phenols extracted by the three methods were subjected to cellular(Ha Ca T)anti-oxidation experiments,the anti-oxidation of three kinds of free phenol cells are as follows:fermentation free phenol>enzymatic hydrolysis free phenol>ultrasonic free phenol;When used Hemerocallis Fulva flower fermentation free phenol extraction in emulsions,it has better stability and skin feel,and is less irritating to human skin compared with commercially available emulsions.Efficacy evaluations in vivo experiments show that emulsions containing Hemerocallis Fulva flower fermented free phenol have better moisturizing properties,and can play a certain role in improving skin dark yellow,increasing skin elasticity and improving skin fine lines,and has good antioxidant effects.
