Poria cocos,also known as Yuling,is the main raw material of traditional Chinese medicine.It belongs to the same food as medicine and food.It can be used as health food.Its main bioactive component is Poria cocos polysaccharide.Poria cocos polysaccharides include water-soluble polysaccharides and acidic polysaccharides.Poria cocos residues are mainly acidic polysaccharides that are difficult to dissolve in water,which are not easy to be directly absorbed by the human body.Most of them are discarded in production,resulting in the inability to make the best use of everything.In order to solve the above problems,taking Poria cocos residue as the main raw material,this thesis studies the extraction process,structural characterization and biological activity of Polysaccharides from Poria cocos residue,and prepares the obtained Poria cocos polysaccharide into a solid beverage,so as to provide a theoretical basis for the further development of Poria cocos and the effective comprehensive utilization of Poria cocos residue.The following are the main research contents and conclusions of this thesis:1?Taking Poria cocos as raw material,the process of water extraction of Poria cocos polysaccharide(hereinafter referred to as water extraction of Poria cocos polysaccharide)was optimized by response surface methodology.The results showed that the extraction rate of Poria cocos polysaccharide was the highest,1.83%,when the time was 3h,the extraction times were 2 times,and the solid-liquid ratio was 1:40.Taking the residue of Poria cocos polysaccharide extracted by water as raw material,the polysaccharide in Poria cocos residue was extracted by enzymatic hydrolysis(hereinafter referred to as enzymatic hydrolysis Poria cocos polysaccharide).The results showed that when the temperature was 55?,pH 5,time was 130 min and the amount of enzyme was 6%,the extraction rate of Poria cocos residue polysaccharide was the highest,which was 8.69%.2?Poria cocos polysaccharide was purified.The molecular weight and monosaccharide components of enzymatic Poria cocos polysaccharide were detected by HPLC,HPSEC and other instruments.The research showed that the relative molecular weight of enzymatic Poria cocos polysaccharide was 1.5-8.5 kDa,mainly containing glucose and mannose.The infrared spectra of water extracted Poria cocos polysaccharide,enzymatic Poria cocos polysaccharide and four kinds of Poria cocos polysaccharides sold in the market were analyzed and compared by infrared spectrometer.The results showed that the two Poria cocos polysaccharides had obvious characteristic absorption peaks,and the similarity between enzymatic Poria cocos polysaccharides and four Poria cocos polysaccharides was 94%-96%.3?the anti-tumor and immunological activities of Poria cocos polysaccharides were studied by CCK-8 method.The proliferation inhibition effect of water extract and poria cocos polysaccharide on breast cancer,liver cancer and gastric cancer cells were evaluated respectively.The results showed that the water extract of Poria cocos polysaccharides had inhibitory effect on the proliferation of hepatoma cells,but the inhibition effect was limited,and the proliferation of breast cancer cells was inhibited.The inhibitory effect of IC50=1174 g/mL on gastric cancer cells was the best,IC50=1096g/mL.Enzymatic Poria cocos polysaccharide had no inhibitory effect on the proliferation of three tumor cells.Both water extracted Poria cocos polysaccharide and enzymatic hydrolyzed Poria cocos polysaccharide can increase mouse spleen lymphocytes,and the effect is different according to the sample concentration and dosage.At the same dose,the effect of water extraction of Poria cocos polysaccharide was stronger than that of enzymatic hydrolysis of Poria cocos polysaccharide;Both water extracted Poria cocos polysaccharide and enzymatic Poria cocos polysaccharide had no side effects on macrophages and had no significant effect on the expression of inflammatory factors.4?Poria cocos polysaccharide solid beverage was prepared with enzymatic hydrolysis of Poria cocos polysaccharide as the main raw material.The auxiliary materials were determined as follows:orange powder 1.65 g,anhydrous citric acid0.054 g,DL malic acid 0.02 g,sucralose 0.009 g,Poria cocos polysaccharide 0.6 g and sorbitol 0.6 g.
