The active substances of Dendrobium officinale include polysaccharides,flavonoids and alkaloids,among which polysaccharides are the active ingredients with the highest content.In recent years,Dendrobium officinale extracts are often used as natural cosmetic raw materials.In the application of Dendrobium officinale,the role of polysaccharide as its main active ingredient is very important,and the biological activity of polysaccharide has a great relationship with the molecular weight.In this thesis,Dendrobium officinale polysaccharides with different molecular weights were used as the research objects to study the effects of Dendrobium polysaccharides in whitening and moisturizing,and to explore the relationship between the molecular weight of Dendrobium polysaccharides and the whitening and moisturizing effects.It is planned to lay a good foundation for the effective application of Dendrobium polysaccharide in whitening and moisturizing cosmetics.The main research contents and results are as follows:(1)Preparation and characterization of Dendrobium polysaccharides with different molecular weightsDendrobium polysaccharides were extracted by water extraction and alcohol precipitation,and 6 kinds of DOPs with different molecular weights were obtained by ethanol stepwise alcohol precipitation and H2O2 oxidative degradation,HPGPC and other analytical methods to compare Dendrobium polysaccharides with different molecular weights,the results show that the two methods of stepwise alcohol precipitation and oxidative degradation will not affect the characteristic groups of Dendrobium polysaccharides.The results showed that the stepwise alcohol precipitation and oxidative degradation did not affect the characteristic groups of Dendrobium polysaccharides,and the structural characteristics of the six different molecular weight Dendrobium polysaccharides did not change significantly due to different molecular weights.(2)Study on the whitening efficacy of Dendrobium polysaccharides with different molecular weightsThe B16 mouse melanoma cell model and the Ha Ca T cell H2O2 oxidative damage experimental model were used to evaluate the effects of different molecular weight Dendrobium polysaccharides on melanin production and tyrosinase activity,as well as the repair and protection of H2O2 oxidative damage.The results show that Dendrobium polysaccharide has the effect of inhibiting melanin production at the cellular level,and within a certain molecular weight range,the larger the molecular weight of Dendrobium polysaccharide,the lower its tyrosinase activity and melanin content.Dendrobium polysaccharide can achieve whitening effect by inhibiting the activity of tyrosinase and reducing the synthesis of melanin in cells.Different molecular weight Dendrobium polysaccharides have protective and repairing effects on Ha Cat oxidatively damaged cells,and the repairing effect is better than the protection effect.Within the experimental range,the protective and repairing effects of medium molecular weight Dendrobium polysaccharides(7.27 kDa-62.55 kDa)against oxidative damage were better.(3)Research on the moisturizing efficacy of different molecular weight Dendrobium polysaccharidesThe moisturizing effects of different molecular weight Dendrobium polysaccharides were evaluated by in vitro weighing method and Ha Ca T cell drying damage experimental model.The results of in vitro weighing experiments showed that in a short period of time,Dendrobium polysaccharides with larger molecular weights had better moisturizing properties;within the experimental conditions,after 24 h,polysaccharides with medium molecular weights had better moisturizing effects than other polysaccharides.The results of cell experiments showed that Dendrobium polysaccharides with different molecular weights could reduce the damage to epidermal cells caused by drying.Within the experimental range,Dendrobium polysaccharides with molecular weights of 62.55 k Da and 4707.98 k Da had stronger resistance to drying damage.(4)Development and performance evaluation of Dendrobium polysaccharide whitening and moisturizing cosmeticsBased on the basic formula,the sensory evaluation was used as the index,and the formula and its process were preliminarily determined by the fuzzy evaluation method,and the preliminary evaluation of moisturizing and whitening effects was carried out on the prepared product.The evaluation results of moisturizing efficacy of whitening moisturizing cream by in vitro weighing method show that whitening moisturizing cream has good moisturizing activity,and the greater the amount of Dendrobium polysaccharide added,the better moisturizing effect.Within the test range,the product with the addition of 1%Dendrobium polysaccharide has the best in vitro moisturizing effect.The evaluation results of the whitening efficacy of whitening and moisturizing cream by in vitro tyrosinase method show that the whitening and moisturizing cream has a good whitening effect,within the test range,when the addition amount of Dendrobium polysaccharide is 1%,the effect of inhibiting tyrosinase activity in vitro is better,and the product has the best in vitro whitening effect.
