Cancer is a serious threat to human health and the occurrence and development of prostate cancer is on the rise year by year.This study aimed to design and prepare dual-antibody immunoliposomes that can engage T lymphocytes and direction their activities towards PSMA-positive cancer cells.Specifically,we tried to conjugate single chain Fv fragments of antibodies against CD3 and prostate cancer cell specific membrane antigen PSMA on liposomes,and optimized their respective densities and ratios on each liposome surfaces.They were then mixed with human T cells and then added into PSMA positive cancer cells.We showed that liposomes were able to activate T cells from human immune system,mediate their interaction with PSMA-positive prostate cancer cells,and eventually effectively kill cancer cells by T cell mediated cytotoxicity.We have demonstrated in this study the feasibility of using dual antibody liposomes as T cell engagers against cancer cells.Further studies are needed in order to optimized the liposome design and improve their activities by incorporating drugs into the liposomes. |
