Objective This topic is to construct a novel multi-functional erythrocyte membrane biomimetic nano-drug targeting lung cancer,which loaded with two chemotherapeutic drugs(paclitaxel and doxorubicin)and investigated its characteriation in vitro and anti-tumor effect in vivo.Methods The self-assembly methoxy poly(ethylene glycol)-poly(lactideco-glycolide)(m PEG-PLGA)nanoparticles encapsulated with paclitaxel(PTX)served as the nano-core of PP-PTX@DRF.Red blood cell(RBC)membranes were loaded with doxorubicin(DOX)periphery to the nanoparticles and the cell membrane surface following the nanoparticle encapsulation was modified with folic acid(FA),then PP-PTX@DRF were successfully constructed.The products were characterized by dynamic light scattering(DLS),transmission electron microscopy(TEM)and protein electrophoresis.The stability of the product was tested by the stability experiment.Lewis lung adenocarcinoma cell line was used for in vivo and in vitro experiments to detect the targeting effect,cell killing effect and tumorigenesis inhibition effect of PP-PTX@DRF.Results Nano-drug had a typical "core-shell" structure,with uniform particle size and hydrated particle size of about 70 nm,which met the requirements of solid tumor Enhanced permeability and retention(EPR)effect.The properties of the protein on the surface of the cell membrane did not change,which ensured the long circulation of the drug in vivo.The stability test showed that it had good stability.PP-PTX@DRF had obvious targeting performance and an enhanced anti-tumor effect on Lewis lung cancer cells in vitro cell experiments.In vivo tumorigenic inhibition experiment showed that PP-PTX@DRF had significantly better tumor inhibition than the original chemotherapy drugs,and had no obvious toxic and side effects.Conclusion A novel multi-functional nano-drug with bionic erythrocyte membrane was successfully prepared,which can load two chemotherapeutic drugs at the same time and exert the synergistic effect of the two chemotherapeutic drugs.The outer envelope of the erythrocyte membrane makes long circulation of nano-drug.With long circulation in vivo and targeting effect increase the drug concentration of tumor tissue,the drug had enhanced tumor killing effect,and lower side effects.
