Nowadays,cancer has become the second-largest death causing disease to humans and about 89.274 million people die because of cancer every year.From 2006 to 2016,the number of deaths increased by 17.8%per year.The development of new medicines to treat cancer is the main focus of this scientific research work..Polyoxometalate has good biocompatibility and significant anti-tumor activity.In our work,the synthesis and anti-tumor activity of polyoxometalate(POM)-based inorganic-organic hybrid crystalline material were studied.The main contents and conclusions are as follows1.Crystalline material 1(Na5[Mn3Na3(trz)7Mn2(H2O)6(SbW9O33)2])was synthesized by introducing 1,2,4-triazole and manganese ions into the trivacancy Keggin-type β-[SbW9O33]9-,trz=1,2,4-triazole.To investigate the in vitro therapeutic efficacy of crystalline material 1,human hepatocellular carcinoma cell lines HepG2 and SMMC-7721 were treated with 1 for 48 h,and cell viability was examined by MTT assay,The IC50 values of 1 on two cell lines were 35.540 ± 0,15 μM for SMMC-7721 and 87.936± 0.13 μM for HepG2,respectively.The binding properties of the crystalline material 1 to calf thymus(ct)DNA were studied using UV/vis spectra.The interaction between the crystalline material 1 and DNA generally leads to a significant change in absorbance with hyperchromism and violet shift of the band.The DNA gel electrophoresis proved that crystalline material 1 could cut the DNA.2.Copper ions and 2,2’-bipyridine were introduced into the[MnMo9O32]6-solution(pH=2.0)to obtain crystalline material 2(H2[Cu6(H2O)(bipy)6(MnMoi2O40)(Mo6O22)]).In vitro assay,the proliferation of hepatocellular carcinoma cells SMMC-7721 was effectively inhibited by crystalline material 2 and the IC50 of crystalline material 2 was 31.69 ± 0.13 μM.Moreover,crystalline material 2 displayed low toxicity towards normal human renal epithelial cells HEK293T and possessed good biocompatibility.3.Crystalline material 3(H2[{Cu(bpy)2}2(MnMO12O40)]·H2O)was synthesized by introducing copper ions and 2,2’-bipyridine into the[MnMo9O32]6-solution(pH=2.4)The cytotoxicity assay demonstrated that crystalline material 3 could effectively inhibit the growth of SMMC-7721 tumor cells,and the IC50 of crystalline material 3 was 31.21±0.37 μM.Besides,The cytotoxicity of crystalline material 3(100 μM)towards SMMC-7721 cells was 0.98 times that of cisplatin.These results proved that the anti-tumor activity of polyoxometalates could be enhanced by introducing organic ligands.
