| The polysaccharides from Pleurotus djamor have many biological activities,such as anti-oxidation,anti-aging,anti-cancer,anti-inflammation effects.The P.djamor mycelia polysaccharides(MPS)were phosphorylated and separated to obtain the phosphorylated MPS component 1(PMPS)and the structure were analyzed.Adenine-induced mouse model of chronic renal failure(CRF)was established to explore the antioxidant,anti-inflammatory and anti-fibrosis effects of PMPS.The experiment results were as follows:(1)The optimal phosphorylation condition was solid-liquid ratio 1:4,STPP:STMP=6:1,reaction time 6h,reaction temperature 80℃,p H=9 and the final phosphate content was 15.22±0.37%.DEAE-52 cellulose anion exchange chromatography was used to separate and purify the components of MPS after deproteinization,depigment and phosphorylated modification.Four components were obtained,and the component with the most polysaccharide content was PMPS.(2)Antioxidant activity in vitro test showed that scavenging DPPH,superoxide anion radicals abilities and reduce power were 52.38±1.27%,57.85±1.56%,and 0.581±0.015,respectively,indicating that the antioxidant activity in vitro of PMPS had the characteristic of dose dependence.(3)Molecular weight,FT-IR,monosaccharide composition,one-dimensional 13C NMR and 1H NMR results showed that PMPS belonged to a homogeneousα-pyranose and was composed of galacturonic acid and glucose with mass fraction of 13.01%and 85.82%.(4)Superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and catalase(CAT)activities of PMPS group were increased and malondialdehyde(MDA)level was decreased when compared with the model(MC)group.Western blotting result showed that PMPS could up-regulate HO-1 protein expression.In addition,PMPS could degrade creatinine(CREA),uric acid(UA)and urea nitrogen(UREA)contents and creatinine kinase(CK)activity,while Hematoxylin/eosin and Apoptotic-hoechst staining suggested that PMPS could effectively relieve renal injury.Moreover,the levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were down-regulated in PMPS groups,and q RT-PCR results also showed the m RNA levels of them were decreased when compared with MC group.Besides,Masson’s Trichrome and immumohistochemical staining results showed that TGFβ1 and Smad3 of the PMPS groups were significantly lower than that of MC group.In summary,PMPS had good antioxidant,anti-inflammatory,anti-fibrosis activities and kidney protection effect on the of adenine-induced CRF mice,which provided a new approach for the development of natural drugs for the treatment of this disease,and also built a foundation for the study of the structure-activity relationship of PMPS. |
