Study On The Antioxidant And Molluscicidal Activities Of Seeds Kernel Extracts From Chimonanthus Praecox Cv.Luteus

Abstract/Summary:
Plants are rich in secondary metabolites,which have a variety of biological activities such as antibacterial,anti-inflammatory,antioxidant and immune regulation,and can be used in the development of medicines,health products and cosmetics.Chimonanthus praecox(L.)Link belongs to the genus Calycanthaceae,Chimonanthus Lindl.It is a traditional garden flower and landscaping plant in country.Ch.praecox leaves,etc.can be used as a single prescription,which has antipyretic and analgesic,cough and phlegm reduction and blood pressure reduction effects,and is used to improve immune system function and treat colds.Ch.praecox cv.Luteus,a variety of C.praecox,is a popular tree species for landscaping.Its inner and outer perianth parts are pure yellow,with rich fragrance.In this paper,Ch.praecox cv.Luteus,which has traditional medicinal value,is selected as the research object,and the extraction,separation and functional activity of its seeds kernel are systematically discussed.The main research results are as follows:1.Antioxidant study of extract from the seeds kernel of Ch.praecox cv.LuteusThe seeds kernel of Ch.praecox cv.Luteus were extracted with 70%ethanol,and the extracts were subjected to gradient extraction with petroleum ether,ethyl acetate,and water-saturated n-butanol to obtain extracts with different polar solvents.The contents of total phenols and total flavonoids in the ethanol extracts and their different polarity extracts of the seeds kernel of Ch.praecox cv.Luteus were determined.DPPH radical scavenging and ABTS radical scavenging were used to evaluate their antioxidant capacity.The results showed that the contents of total polyphenols and flavonoids in ethyl acetate extract were the highest,which were 6.16 mg/g and 8.79mg/g,respectively.The ethyl acetate extract had the strongest ABTS radical scavenging activity with IC50 value of 28.35 mg/m L,followed by ethanol extract(IC50=42.75 mg/m L).The ethyl acetate extract had the best DPPH radical scavenging activity with IC50 value of 9.67 mg/m L.2.Extraction,separation and structural identification of volatile components from the seeds kernel of Ch.praecox cv.LuteusCh.praecox cv.Luteus seeds kernel were extracted by heating with n-hexane,and the n-hexane extract was saponified to obtain total fatty acids and unsaponifiables.Gas chromatography-mass spectrometry(GC-MS)was used to analyze the seeds kernel of Ch.praecox cv.Luteus.The chemical composition of total fatty acids and unsaponifiables were analyzed.A total of 16 fatty acids were detected from in the seeds kernel of Ch.praecox cv.Luteus,13 of which qualitatively identified,the content of octadecadienoic acid(56.76%)and palmitic acid(17.03%)was the highest;19 unsaponifiable compounds were identified,the main components of which were tricosane and eicosan.3.Study on the molluscicidal activity of the volatile components from the seeds kernel of Ch.praecox cv.LuteusTo detect the killing activity of volatile components of Ch.praecox cv.Luteus seeds kernel on Pomacea canaliculata,to evaluate the degree of liver injury and the physiological and biochemical effects of the soaking treatment of volatile components of Ch.praecox cv.Luteus seeds kernel on P.canaliculata.The results showed that after 48 h soaking in fatty acid of Ch.praecox cv.Luteus seeds kernel,the hepatopancreas of P.canaliculata had a large area of necrosis.The contents of soluble sugar,soluble protein,and albumin(Alb)in the hepatopancreas of P.canaliculata decreased with increasing FKC concentration.The content of malondialdehyde(MDA)and the activities of cereal third transaminase(ALT),aspartate transaminase(AST),alkaline phosphatase(AKP),and acetylcholinesterase(ACh E)increased with increasing FKC concentration.The results showed that FKC has an obvious negative effect on the hepatopancreas cell structure and physiological function of P.canaliculata,i.e.,has strong molluscicidal activity.
