At present,there are two problems in Chinese fermentation industry:(1)With the increase in the quantity and complexity of fermentation data,the problems of fermentation data management and effective utilization have gradually prominent;(2)Low degree of automation in the fermentation industry,especially for large-scale fermentation enterprises,which rely on skilled workers,and the output and quality of products fluctuate greatly.In order to solve these problems,this project uses Django framework and MySQL and MongoDB database software and other tools to develop the Bio JN fermentation technology cloud server program.On the basis of solving the management and utilization of fermentation data,taking Pichia pastoris as the research object,sharing the process control software package through the Internet,and realizing automation in the key link of the high-density culture of Pichia pastoris.Below are key research findings:(1)API interface development:The server program is designed with full consideration of the user’s various usage scenarios,and a rich variety of API interfaces are reserved for this to meet the user’s demands.The research room has developed a PC client program in the early stage,users can use the PC client program to access the API provided by the server to realize data management,data calculation,parameter configuration,and process software package download.(2)Development of a generic fermentation database:A general fermentation database system was designed by using MySQL(relational database)and MongoDB(non-relational database).Online parameters with strong regularity and strict format are stored in MySQL data table.It is very arbitrary to save in MongoDB documents,and lack of offline measurement parameters and event records in strict and unified data format.(3)Process control software package development:Based on the long-term research of the Pichia pastoris fermentation process proposed by the author’s research team,strategy A:modified DO-Stat glycerol feeding strategy,strategy B:"limited methanol concentration-sufficient dissolved oxygen concentration"and strategy C:"methanol"Adequate concentration-limited dissolved oxygen concentration",the mechanism of these strategies was analyzed,and these control strategies were compiled into a Pichia pastoris process control software package.(4)The Mut+type strain expressing HSA-GCSFm and the MutS type Pichia pastoris expressing hLYZ were used as model strains to verify the above-mentioned process control software package performance.The results showed that:Mut+type Pichia pastoris induced 60h,the concentration of HSA-GCSFm reached 532 mg·L-1;the average hLYZ total enzyme activity arrived 84 032 U·m L-1 after the induction of Pichia pastoris MutS for 69 h.The concentration/titer of the above two target proteins reached a high level,separately using the Mut+type pichia pastoris production of porcine circovirus virus type Cap protein and the pichia pastoris producting pig interferon-alpha(pIFN-alpha)in the process of using the network sharing strategy to fermentation,The results showed that the concentration of Cap protein reached 0.17 g·L-1 and the concentration of pIFN-αreached 1.78 g·L-1.It shows that this system has high foreign protein output and good versatility after use,and it verifies the feasibility/repeatability of this network sharing system.
