Raman scattering spectroscopy is a very powerful analytical technique,which has been widely used in biochemistry,medicine,petrochemical industry and other fields in the past decades due to its high sensitivity,high speed and noninvasive advantages.However,the original Raman spectrum usually contains noise and fluorescence background interference components,which hinders the application of Raman spectrum analysis,so the removal of fluorescence background is very important data processing step.In order to make better use of Raman spectroscopy technology to carry out application research,it is particularly important to develop a successful de-background system software with high operating efficiency.This paper mainly focuses on Raman spectral background algorithm research and system design.The following is a brief overview:In this paper,an improved algorithm for improving Poly Fit Raman spectral fluorescence background(Ipro-poly)is proposed.This algorithm inherits the advantage of polynomial fitting and improves the algorithm based on the Raman spectrum characteristics of biological sample of interest to our group.Algorithm of original spectral ordinate values compared with each wave number of the polynomial model,for each round continuous polynomial fitting,select each lower limit value of wave number,then together constitute a fixed spectrum,fitting,again based on the polynomial order number at the right moment,through the multiple iterative fitting out the background of a smooth curve.The algorithm also considered the distortion of signal noise and the influence of large noise,combined with the de-peaking process added in the first iteration,so as to minimize the interference of human to the background spectrum deduction work,and obtained the most representative pure Raman spectrum.The software system designed in this paper is easy to operate,easy to understand,and has a more humanized humanmachine interaction interface.The software implements four algorithms to process the original Raman spectral data,and can display the processed data in the preset subinterface of the system and save them according to the actual needs.At the same time,the software also has the normalization function in the preprocessing process of Raman spectral data.Based on the effective operation of Ipro-poly algorithm,this paper develops the software of Raman spectrum background removal system,which solves the interference problem of fluorescence background to the original Raman spectrum signal,and connects the whole Raman spectrum data preprocessing process in series,which makes the data preprocessing process convenient and fast,and provides a reliable data source for the later analysis of Raman spectrum data.
