Specific proteins,are some proteins derived from tissue cells,widely found in the serum containing specific functions of proteins,many diseases can cause changes in serum proteins,specific proteins have become an important clinical test indicators.Protein has good antigen,can use antigen-antibody reaction to determine a protein with a specific antigen,so it is called specific protein detection.The detection of proteins or chemicals such as lipid carrying proteins,trace albumin,creatinine and creatine kinase in neutrinocytes is of great significance to the diagnosis of early kidney injury,diabetic nephropathy and cardiomyopathy.In this paper,using Lambeth-Bill’s law as the basic detection principle,a fully automatic protein-specific detection device for the above-mentioned substance detection is designed and developed.Using the photoelectric effect of photoelectric sensors,the device uses the turbidity method or color ratio method to detect the material content in the sample quantitatively by the method of relative or absolute absorbance of the sample.The device mainly includes optical system design,liquid system design,mechanical structure design,hardware circuit design and embedded programming,etc.The performance verification experiment results show that the automatic protein detection device built according to the design scheme described in this paper has good non-biological performance and biological performance.The maximum current difference of the constant current source of the light source is 7 m A,the error of 100μL liquid quantity is less than 4μL,the temperature control stability of the constant temperature region is±0.1℃,the output stability of the signal detection unit is±10 m V,and the maximum error of the repeated motion of the 3D motion platform is 76.667μm.The linearity of the device in the detection of neutrophil delivery protein(NGAL),trace albumin(MALB),creatinine(CRE),creatine kinase(CK)and other biochemical items was good,and the R~2 was 0.990,0.987,0.987,0.989respectively.Compared with Hitachi 7180 automatic biochemical analyzer,the detection results showed high concordant degree.The negative concordant rate of albumin(ALB)was92.06%,and the positive concordant rate was 89.36%,and the negative concordant rate of ALB/Cre was 87.18%,and the positive concordant rate was 92.96%.The parameters of the automatic specific protein detection device built according to the scheme described in this paper meet the detection requirements of neutrophil gelatase-associated lipid carrier protein,trace albumin,creatinine,creatine kinase and other biochemical indicators.The results of this study provide an experimental basis for the development of automatic detection device for specific proteins.
