| Through single factor experiment,the experiment explored the influence of response time,the pH value of reaction solution,chitosan-bentonite ratio and reaction temperature,on bentonite-chitosan compolexes(Bt-CTS compolexes),optimized preparation conditions by orthogonal experiment,and compared the adsorption efficiency of fumonisin B1 by Bt-CTS compolexes in different media.The study provided references for developing the adsorbent of fumonisin B1.Expt.l.Effect of different preparation conditions on Bt-CTS compolexesThrough single factor experiment,the experiment explored the influence of response time(5,10,15,30 and 60 min),the pH value of reaction solution(0,0.5,1,3 and 5),chitosan-bentonite ratio(0.1,0.2,0.4,0.6,0.8,1.0 and 1.6)and reaction temperature(60℃,70℃,75℃,80℃,85℃ and 95℃)on Bt-CTS compolexes.The experiment made chitosan content of Bt-CTS compolexes as an indicator of compolexes’ production effectiveness,and plotted the graph of pH-time curve during reaction process of chitosan mixed bentonite suspension,and compared the solubility of chitosan in hydrochloric acid with different pH value.The result showed that with the condition of response temperature 60℃,chitosan-bentonite ratio 0.8,and pH value 1,the response bewteen chitosan and bentonite reached the state of equilibrium,and the optimum pH value of reaction was 1,and the best chitosan-bentonite ratio was 0.6,and the optimal reaction temperature was 85℃.Expt.2.Optimized preparation conditions ofBt-CTS compolexesOn the basis of the preliminary results of trail 1,the study took response time(10 and 15 min),the pH value of reaction solution(0.5 and 1),chitosan-bentonite ratio(0.4 and 0.6),chitosan’s degree of polymerization(poly-chitosan and oligochitosan)and response temperature(85℃ and 95℃)as factors of L16(25)orthogonal experiment design method,futher optimized preparation conditions of Bt-CTS compolexes.The experiment elected the optimal preparation conditions taking chitosan content of Bt-CTS compolexes and adsorption efficiency of fumonisin B1 as indexes.The results showed:(1)taking chitosan content as an index,optimal preparation conditions were:response time was 10 min,the pH value of reaction solution was 1,chitosan-bentonite ratio was 0.6,response temperature was 85℃ and oligochitosan was better effectiveness;(2)taking adsorption efficiency of fumonisin B1 as an index,optimal preparation conditions were:response time was 15 min,the pH value of reaction solution was 0.5,chitosan-bentonite ratio was 0.6,response temperature was 95℃ and poly-chitosan was better effectiveness.Expt.3.Evaluation of binding efficiency of fumonisin B1 by Bt-CTS compolexes in different mediaTaking purity water,the superate of gastric and intestinal chyme of pigs as reaction media,the adsorption efficiency of fumonisin B1 by Bt-CTS compolexes were evaluated.The result showed that the adsorption efficiency of fumonisin B1 in purity water was optimal,following in the supernate of gastric chyme,and the supernate of intestinal chyme worst.In purity water,the adsorption efficiency of fumonisin B1 by the third and the fifth Bt-CTS compolexes achieved more than 85%,but the adsorption efficiency of fumonisin B1 was less than 30%. |
