| Trichome is a subsidiary structure of epidermal cells,which plays an important role in defense protection and phase transition.At present,the development characteristics and molecular mechanisms have been extensively reported about single epidermal cell differentiation trichome in Arabidopsis thaliana.There is few related article in rice.In this study,the rice leaf trichome of seedling,tillering,and booting stages were observed to induce the distribution character of trichome during full growth period by means of Scanning Electron Microscopy(SEM).The exploration of glabrous gene OsSPL10 responsed to exogenous hormone and related genes function by analyzing 3 glabrous mutants sst,oswox3b,and hl6,which could establish foundation of trichome development characteristic and molecular mechanism in rice.The main results are listed as follows:(1)The trichome development characteristics were analyzed about different parts and axial surfaces in the whole growth period,which summed up that trichome development started in early stage leaf growth and trichome development mature was completed in the advance stage of growth.The trichome development program in leaf parts was closely connected with the establishment of three-dimensional axis polarity.The development of macro-hair was earlier than micro-hair and glandular,and the macro-hair number was fewer.These results showed that the mature development of rice trichome was related to the rice growth period.(2)The relative expression levels of OsSPL10 in OE plants were ten to several dozen times than control,and the number of macro-hair was significantly increased.Combination with the glabrous phynotype of mutant sst,we conclude that OsSPL10 positively regulated macro-hair development.(3)The expression of OsSPL10 could be induced by exogenous IAA,GA3 and MeJA in wild type R401,and the mutant sst was sensitive to GA3 and MeJA,was insensitive to IAA.The mutation of OsSPL10 affected the expression of GA signaling genes(OsKAO,OsGA20ox2 and OsGIDl)and JA signaling genes(OsLOX2,OsJAR1 and OsCOI1).Howerver,the correlation both OsSPL10 and JA signaling pathway was higher than GA.The results suggestted that GA and JA signaling transductions were mediated by OsSPL10 gene during them involved in the reglation of trichome development.(4)The mutant oswox3b was obtained by CRISPR/Cas9 technology,and the leaf and mature seed trichome of mutant was significant degradation,particularly in macro-hair.The expression level analysis of OsSPL10,OsWOX3B and OsPLT2 gene in glabrous mutants sst,oswox3b and hl6 indicated that OsSPL10 acted the upstream of OsWOX3B and OsPLT2,and positively regulated their expression.(5)Protein OsSPL10 was physical interaction with OsWOX3B and OsPLT2 respectively by co-transformation of yeast two-hybrid system in vivo.Both OsSPL10 and OsWOX3B protein,OsSPL10 and OsPLT2 protein could interact in tobacco cell by bimolecular fluorescence complementation.The results showed that there were interaction between OsSPL10 and OsWOX3B,and OsSPL10 and OsPLT2 respectively. |
