| It is estimated that Up to 10% of losses in aquaculture animals are due to infections,causing economic losses more than $10 billion in the global.It is essential that prevent and control disease for sustainable aquaculture.Prevention measures based on stimulating the fish immune system have become the key to the successful development of fish culture.Photobacterium damselae subsp.Damselae stored in laboratory,The whole formalin inactivated vaccine was prepared,and the Cynoglossus semilaevis was immunized by soaking and injecting.Four tissue samples of liver、kidney、gill and spleen of experimental fish were taken at nine time points: 0h、4h、8h、12h、24h、48h、72h、96h、7d after immunization.Using fluorescence quantitative method to detect the experiment of fish in the different time points after immune four tissues of TLR2、My D88、(NF)-κB、CD28、CD40、(IL)-1β、(IL)-8、(IL)-6、CXCe1 and(TNF)α the 10 kinds of immune-related genes expression changes.The study showed that:1.Among the two immune methods,except that the expression of IL-1β in liver and kidney and My D88 in liver of immersion group were lower than that of control group,the expression of other genes in all organs increased compared with blank group.2.The expression levels of the remaining genes were up-regulated in all tissues compared with the control group.The expression level of most genes in the immersion group was significantly higher than that in the control group at 0h of immune treatment.In general,the highest expression level of each tissue in the immersion group occurred at 24h-7d,and the maximum expression level of each gene was 2-15.65 times that of the control group.The highest expression level of each tissue in the injection group appeared at 4-48 h,and the maximum value of each gene expression was 2-21.51 times that of the control group.3.In the two immunization methods,except that the peak time of TLR2 expression in the gill tissue and CD28,IL-1β,and TNF-a expression in the kidney of the immersion group was earlier than that of the injection group,the peak time of gene expression in the immersion group was later than that of the injection group. |
