Andrographis herba is derived from parts of Andrographis paniculata(Burm.F.)Nees belongs to Acanthaceae family,bitter and cold,returning to the lungs,stomach,large intestine,and small intestine meridians.It is used as medicine above ground,with heat-clearing and detoxifying,cooling blood,and swelling Efficacy,used for cold and fever,sore throat,sore tongue,cough,diarrhea and dysentery,hot astringent pain,carbuncle sore,snake bite,etc..More than 99%of China’s andrographis commercial medicinal materials are derived from cultivated species,and only very few are derived from wild species.Due to long-term artificial cultivation management,the plant currently has a single germplasm,the quality of medicinal ingredients has declined,and the quality is The problem of stability seriously affects the quality and sustainable development of andrographis paniculata.Therefore,it is imperative to conduct research on selection and breeding of new varieties of Andrographis paniculata.Irradiation mutation breeding is the use of gamma rays,X-rays or other physical mutagenesis factors to irradiate plants to obtain new varieties with genetic mutations in a short time,so that people can screen out new varieties with excellent traits.Emerging breeding technologiesIn this study,60Co-γ rays(0,10,20,50,100,200,300Gy)with different irradiation doses were used to plant andrographis paniculata seeds after irradiation treatment.The botanical characteristics,active ingredient content,physiology The active substance content,stress resistance and CPS gene expression were measured to determine the application value of 60Co-γ ray irradiation in andrographis paniculata mutation breeding technology.The results show1.60Co-γ ray irradiation with different irradiation doses has different effects on the botanical properties of andrographis paniculata.The emergence rate,root length,and number of stomata show a downward trend with the increase of irradiation dose;plant height,leaf area,Fresh weight,dry weight and other indicators showed an upward trend and then a downward trend with the increase of radiation dose.Among them,andrographis paniculata plant height,leaf area,fresh weight,dry weight and other indicators in the 100Gy dose group were the highest,which were 124.37%,238.58%,143.61%,and 119.42%in the control group;plant height,leaf area,The fresh weight,dry weight,and number of stomata were the lowest,which were 48.97%,65.85%,46.64%,31.59%,and 35.88%in the control group,respectively.2.T-SOD enzyme activity and CuZn-SOD enzyme activity increased first and then decreased with the increase of irradiation dose.Among them,the 100Gy dose group had the highest chlorophyll content,T-SOD enzyme activity,and CuZn-SOD enzyme activity,which were 105.31%,101.65%,and 106.75%of the control group,respectively.3.The germination rate,plant height,leaf area,fresh weight and dry weight are not significantly correlated with the irradiation dose,the emergence rate and the irradiation dose have a very significant negative correlation,and the root length is significantly negatively correlated with the irradiation dose.There was a significant negative correlation between the stomatal number and the radiation dose.Regression analysis was performed on the emergence rate to obtain a linear regression equation y=54.784-0.139x(y represents the emergence rate and x represents the irradiation dose),and the LD50 was 195.10Gy.4.Andrographolide is distributed in the above-ground part of Andrographis paniculata,leaves,stems,and roots,and the content is respectively leaf>above part>stem>root,while dehydrated andrographolide is only distributed in the above-ground part,leaves and stems of andrographis paniculata.,The root could not be detected The content was leaf>ground part>stem,and the content of total lactone in andrographis paniculata was leaf>ground part>stem>root.Among them,the content of active ingredients in the aerial part of andrographis paniculata increased to varying degrees with the increase of the irradiation dose.The 20~300Gy dose group can significantly increase the content of andrographolide and dehydroandrographolide.The 50Gy dose group has the highest total lactone content,which is 200.75%of the control group,followed by the 100Gy dose group,which is 155.50%of the control group.;Andrographis paniculata leaves active ingredient content increased to varying degrees with the increase of radiation dose,of which the 50Gy dose group had the largest total lactone content in the control group,which was 130.37%,followed by the 20Gy dose group,which was the control group.The total lactone content in the stem portion of the 10Gy,50Gy,100Gy,200Gy,and 300Gy dose groups was reduced to varying degrees compared with the control group,which were 98.61%,84.75%,99.41%,and 79.41%of the control group,respectively.The content of andrographolide in the root of the 10-200Gy dose group was reduced to varying degrees compared with the control group,which were 77.27%,77.27%,81.82%,54.55%,and 68.18%in the control group,and the 300Gy dose group was compared with the control group.There was a significant increase of 181.82%in the control group5.50Gy of 60Co-γ rays can improve the drought resistance and salt tolerance of Andrographis paniculata seeds.50 Gy of viCo-y irradiation siguificantly promoted the growth of Andrographis paniculata under drought and salt stress,including the promotion of seedling radicle and hypocotyl elongation6.The relative expression of CPS gene in andrographis paniculata leaves increased first and then decreased with the increase of irradiation dose.Among them,the expression of CPS genes in the 50Gy and 100Gy irradiation dose groups were 15187%and 135.08%in the control group,respectively.Correlation analysis between total andrographolide and CPS gene expression in Andrographis paniculata samples from Guangxi Yulin andrographis paniculata treated with different irradiation doses was performed using SPSS software.The correlation coefficient was 0.827,indicating that the expression of CPS gene was significantly positive
