Abstract/Summary:
Pyroapoptosis is a rapid pathological suicide process that relies on caspase-1(classical pathway)and caspase-11(mouse pathway)and 4/5(human pathway)(non-classical pathway)pathways.In the course of severe ulcerative colitis,the intestinal tract of the body in a relatively short period of time appears hematocellular damage symptoms.At present,there are few studies on the mechanism of ulcerative colitis in China.The purpose of this study was to investigate the effect of shenling baizhu powder on the caustic cell death pathway in the treatment of ulcerative colitis.In the first part of the experiment,C57BL/6J mice were given sodium glucan sulfate solution of different concentrations to induce ulcerative colitis.Appropriate concentrations of sodium glucan sulfate were selected by body weight,fecal occulted blood,organ index,blood routine,TNF-?,IL-1?,colon pathological sections,GSDMD-N and NLRP3.The test results showed that there was a positive correlation between the severity of ulcerative colitis and the concentration of sodium glucan sulfate,and the blood stool and weight loss in the 4% group and the 5% group were serious.In the second part,mice were given 3% dextran sulfate sodium for 7 days to induce ulcerative colitis,and shenlingbaizhu powder and melalazine with different concentrations were given for 7 days respectively.The effects of shenlingbaizhu powder and melalazine with different concentrations on colitis were observed through indicators such as weight,survival rate,fecal occultum blood and inflammatory factors.Results: the survival rate of mice in the blank control group was 100%,55.5% in the model group,66.5% in the low-dose Chinese medicine group,70% in the medium-dose Chinese medicine group and the high-dose Chinese medicine group,and 25% in the western medicine group.The DAI score of mice with ulcerative colitis was significantly increased by shenlingbaizhu powder,which increased the weight reduced by colitis.To alleviate the colonic shortening caused by ulcerative colitis in mice;It can effectively reduce the white blood cell increase caused by ulcerative colitis and increase the red blood cell decrease caused by hematoecia in mice.The serum levels of TNF-?,IL-1?,IL-18,caspase-1 and caspase-11 increased by colitis were effectively reduced.Conclusion: shenlingbaizhu powder can effectively reduce the expression of gsdmd-n,the key protein of pyroptosis in colitis and colon tissues,and has a certain down-regulation effect on inflammatory protein NLRP3.In the third part of the experiment,10% glucan sodium sulfate was used to stimulate for 2 h,followed by 10μg/m L LPS for 12 h to establish a cellular inflammatory model.TNF-?,IL-1?,IL-18,caspase-1,caspase-11 and other indicators in the supernatant of cell culture were determined to observe the effects of lingbaizhu powder with different concentrations on the cellular inflammatory model.Results: The concentrations of IL-1? and IL-18 in the blank control group were lower than those in the model group and three groups of traditional Chinese medicine with different concentrations(P<0.05).All the three concentrations of TCM were lower than the model group(P<0.05),and there was no significant difference between the three concentrations of TCM(P>0.05).The concentrations of caspase-11 in the model group were all higher than those in the blank control group(P<0.05)and the three traditional Chinese medicine groups.There were significant differences between the 1.25% group,0.62% group and the model group(P<0.05),no significant differences between the two groups(P>0.05),and no significant differences between the three groups(P>0.05).The concentrations of caspase-1 in the model group were all higher than those in the blank control group and the three traditional Chinese medicine groups,and there was no significant difference between the groups(P>0.05).Summary: Shenlingbaizhu powder may reduce inflammation by regulating the classical(caspase-1)and non-classical(capase-11)pathways in pyroapoptosis,in which the non-classical(capase-11)pathway may play a major role.
