The differentiation and development of skeletal muscle is an extremely complex and precise process,involving a variety of acting factors,signal transduction pathways and complex regulatory networks.Therefore,it is very important to understand the basic process of skeletal muscle differentiation and development and the main factors involved in it.In this study,proteomics analysis was carried out on the cell samples of bovine skeletal muscle satellite cells at different development stages by using protein spectroscopy,and bioinformatics analysis revealed changes in protein expression during muscle cell development and screened key differential proteins.In this study,bovine skeletal muscle satellite cells were used as test materials.Total protein samples of bovine skeletal muscle satellite cells during the proliferative phase(GM),the second day of differentiation(DM2)and the third day of differentiation(DM3)were collected,then the proteomic analysis of different stages was completed by protein mass spectrometry.A total of 4196 proteins containing at least two specific peptide segments were identified.The protein data of three different periods were compared and screened by t-test.In DM2 vs GM Group,140 differential proteins were obtained,including 52 down-regulated proteins and 88 up-regulated proteins.In DM3 vs GM Group,222 differential proteins were obtained,of which 101 were down regulated and 121 were up regulated.In DM2 vs DM3 group,36 differential proteins,15 down-regulated proteins and 21 up-regulated proteins were obtained.In General,the differences of proteins in the two developmental stages of proliferation and differentiation were large,and the changes of protein expression in the second and third days of the same developmental stage were relatively small.At the same time,through go and KEGG analyses,it was found that the differential protein was significantly enriched in the biological processes of skeletal muscle contraction and regulation,sarcomere tissue,fast and slow muscle fiber transformation.In the molecular function(MF entry),it was significantly enriched in actin binding,tropomyosin binding,oxygen binding and iron ion binding.The enrichment of KEGG pathway mainly focused on DNA replication,nucleic acid repair,PPAR and other processes.The protein interaction analysis(STRING)found that the three groups of differential proteins can be mainly divided into three major interaction groups of muscle fiber development related proteins,DNA synthesis related proteins and blood transport related proteins,which play the roles of actin binding,tropomyosin binding and DNA synthesis.To summerize,this study used efficient proteomics technology to screen and identify the differential proteins in the proliferation and differentiation of bovine skeletal muscle satellite cells,laying a foundation for in-depth analysis of the mechanism of muscle growth and development and promoting the genetic improvement of beef cattle breed.
