| Paris as a valuable traditional Chinese medicine,the wild resources have been reduced sharply,The Dabie mountainous area is an important natural distribution area of the genus paris and one of the traditional producing areas of paris as listed in the pharmacopoeia.However,the actual original plants of paris are relatively mixed,and the quality of paris is uneven.By infrared spectrum method,this paper use omnic software to identify research of heavy floor plant,using the method of HPLC identify different species 、growth year 、ages and seasons polyphylla saponin content in the paris rhizomes for anhui dabie mountain area,carry out the breeding of new cultivar of paris,The main research contents and results are as follows.(1)The proper temperature of water bath heating and alcohol concentration of 90 % should be80 ℃,and the extraction times should be once.(2)Rhizome drying rate of 4 kinds of heavy buildings and 3 kinds of different years and seasons was between 20%-40%.Among them,the P.fargesii var.Petiolata rate of 6-year rooting in December was the highest(39.03%),the 4-year old folding rate was relatively low(23.02%).(3)Qualitative evaluation of the infrared spectrum of rhizomes of different species and growth years showed that the wavelengths of the two-dimensional derivative chromatographic spectrum at 1800-800 nm contributed more to the differentiation of Paris rhizomes.According to the similarity analysis of the graph,the similarity between P.polyphylla var.Stenophylla and P.polyphylla.var.latifolia is the highest,and the similarity between P.polyphylla var.Stenophylla and P.fargesii var.Petiolata is the lowest.(4)HPLC method was used to determine the content of total saponins in the rhizomes of 4species of the Paris with strong rhizomes about different growth years and different seasonsin Dabieshi Mountain of Anhui Province.The results showed that: In October,the content of total saponins was the highest in 4-year old P.fargesii var.Petiolata,which was 18.60 mg/g(1.86%).In August,the content of total saponins in 6-year old P.polyphylla.var.latifolia was relatively low,which was 1.30 mg/g(0.13%).The results showed that the total saponins contents of paris in the 6-year old rhizomes of the four species at different ages were generally lower in the 1-year,2-year and 3-year segments.while the contents of total saponins in the P.fargesii var.petiolata were higher in the range of 0.63 % to 1.11 %.The content of total saponins in the 6th year section of the P.fargesii var.petiolata rhizome was the highest,which was 14.20 mg/g(1.42%).The content of total saponins in the 6th year segment of P.polyphylla var.stenophylla was relatively low,which was 0.30 mg/g(0.03%).(5)Breeding and registration a new Cultivar about paris,it is No.1,Hui paris with a growth period of 235 days,neat field growth,strong plants,and vigorous growth.The 5-year-old produces an average of 2366.6 kg of fresh rhizomes per mu,with saponin content of 0.80%,which is higher than that of CK(parent)and better than the standard of 0.6% in the Ch.P.The research results can provide guidance for the rational development and utilization of the resources of paris in the Dabie Mountains. |
