Colletotrichum gloeosporioides is one of the most serious fungal diseases of apple,which occurs in all apple producing areas in China.For understanding the incidence of apple anthracnose,12 orchards around Shenyang city were investigated,including Kangping,Liaozhong,Faku and Xinmin County and Hunnan District,Shenbei New Area and Shenyang Agriculture University.We found that the disease in Liaozhong region was relatively heavy and it’s in the Kangping region is relatively light.The pathogenic strains were collected,45 independent strains of anthrax were identified from 78 samples,and the most pathogenic strains were screened by classifying and determining the virulence of different strains,the pathogenicity test showed that strain SL2-3 had the strongest pathogenicity.The indoor bacteriostasis test of using 14 common fungicides showed that Prochloraz had the better effect.In addition,the autotetraploid of ‘Hanfu’ apple was found to be more resistant to anthracnose than ‘Hanfu’,and the expression of MIR390 was up-regulated in the tetraploid by mi RNA Sequencing.This is the first time to clone the precursor sequence of MIR390 from ‘Hanfu’.Four apple transgenic lines overexpressing MIR390 were obtained by agrobacterium-mediated method.After Colletotrichum gloeosporioides treatment,the overexpressing MIR390 transgenic plants had higher anthracnose resistance than the wild-type plants.
