Wild macrofungi are a kind of fungi that form large fruiting bodies among the natural fungi,many of which have edible and medicinal value.At present,there are many related researches on the identification and isolation of wild large fungi at home and abroad,but the applied research in the biological control of plant diseases is rarely studied.Therefore,this experiment relied on the extensive wild macrofungi resources in Mudanjiang area to collect,molecularly identify and organize wild fungi in the four surrounding areas of Mudanjiang.Based on the operation of using concentrated sulfuric acid to regulate soil acid and inhibit disease in rice production and seedling period,the acid producing strains of wild macrofungi collected were screened out according to p H value,and the inhibition effect of wild acid producing strains on Magnaporthe grisea was determined.The test results are as follows:1.A total of 128 species of large fungi fruiting bodies were collected,and 97 fruiting body DNA samples were extracted,and sequenced after PCR using fungal common knowledge primers.After BLAST comparison and phylogenetic tree construction,69 samples were identified and belonged to There are 14 families,27 genera,and 48 species.The dominant family is Agaricaceae and Tricholomaceae,accounting for 16.33 %,and the subdominant family is Russulaceae and Polyporaceae,accounting for 14.29 %;the dominant genus is Agaricus,accounting for 14.58 %,the secondary dominant genera are Agaricus and Russula,accounting for 8.33 %.2.The fruit body tissue was separated and purified at the first time after the sample was collected,and finally 15 isolated cultures were obtained.The mycelial DNA was extracted and sequenced after fungal common primer PCR,and compared with the corresponding original fruit body PCR product sequence,and the final identification was obtained.14 pure cultures of wild large fungi.3.The p H value of the metabolic fluid of 14 wild fungi in liquid culture was measured by pure culture.Among them,the p H value of the metabolic fluid of 3 strains was <4.5,especially the p H value of the A24 strain was as low as 2.5,and 3 strains were selected as spare strains for the Oryza sativa test.The results of the confrontation co-cultivation showed that the three acid-producing wild bacteria had significant antagonistic and inhibitory effects on Magnaporthe grisea.Strain A24 had a very significant antagonistic effect and completely inhibited the melanin secretion of Magnaporthe grisea.Follow-up tests continued on the A24 strain.Design.4.The results of the solid culture antibacterial test showed that the metabolic liquid of A24 strain can change the growth habit of rice blast fungus which produces less aerial hyphae,that is,the hyphae are mainly surface aerial and rarely penetrate into the matrix;the metabolic roll of A24 strain has a very significant inhibitory effect on the melanin secretion of Magnaporthe grisea and the melanin performance of Magnaporthe grisea is blocked when the dosage is greater than 60 %;the results of the liquid culture antibacterial test show: The metabolic fluid of strain A24 has a very significant inhibitory effect on the growth of rice blast fungus;when the added concentration of the metabolic fluid reaches 50 %,it can inhibit mycelial germination and growth.When the added concentration reaches 75 % or more,the rice blast fungus in the culture medium hardly grows;When it is more than 50 %,the rice blast fungus has no melanin secretion.It is proved that a certain concentration of A24 metabolite can effectively inhibit the growth of rice blast fungus and block its melanin secretion.In summary,acid-producing wild fungi have a very good inhibitory effect on the growth of Magnaporthe grisea,and have great application value in the prevention and treatment of Magnaporthe grisea.They can be used as an ideal plant protection product in production.,Provide resource reserves for high-efficiency biological agents in the prevention and control of plant diseases and insect pests.
