Study On Cough Relieving Effect Of Traditional Chinese Medicine Compound "Manzhijie" Oral Solution And Its Curative Effect

The traditional Chinese medicine compound MOS is composed of manshan red,platycodon grandiflorum and zhilicorice root.Its granule is a classic prescription of human medicine,mainly used for the treatment of human tracheitis,cough and asthma,cold and asthma and other common diseases.In this study,aiming at the harm of animal respiratory diseases,a mouse cough model was established to evaluate the antitussive effect of MOS.Taking piglets as clinical test animals,the infectious pleural pneumonia disease model of piglets was established to evaluate the safety and antitussis effect of compound MOS on piglets.Through the above studies,the foundation was laid for the application of MOS in veterinary clinic.The main findings are as follows:1.Effect of MOS on capsaicin-induced mouse cough modelTendency,with 0.1 L-1capsaicin spray method to induce mice cough,through the determination of the trigeminal nerve of mice whether building success,content of substance P in the building before and after the cough,histopathologic autopsy after dosing,through rt-pcr test drug group and model group mice trachea tissues(TRPV1)receptor m RNA expression.The results showed that:the release of substance P in the trigeminal ganglion was significantly increased in the model group compared with the normal group after 3 days of continuous modeling(p<0.05).Compared with the model group,the number of coughs in each group decreased significantly,and there was a significant difference between the low-dose group and the medium-high dose group(p<0.05).MOS(p<0.05).Compared with the model group,each dose group of MOS had an inhibitory effect on the expression of TRPV1m RNA,and the inhibitory effect in the medium dose group was greater than that in the high dose group.Group dose have normal alveolar structure,including low dose group found a slight congestion,lung lobules area dose group found in the alveolar walls capillary hyperemia,high dose group of the capillaries and interstitial blood vessel hyperemia,cavity clean,no leakage,the experimental results show that main orange"with"oral solution to alleviate pathological changes.2.Safety test of target animals(piglets)with oral solution of MOSPiglets were randomly divided into four groups(8 heads in each group):blank control group,low-dose group(0.278ml/kg),medium-dose group(0.834ml/kg),and high-dose group(1.39ml/kg).In the oral solution group of MOS,the dosage was recalculated according to the total population of piglets,and the dosage was continuously administered by gavage for 7 days.The body temperature,heart rate,respiration,blood routine and biochemical parameters of piglets were observed during the experiment.At the end of the experiment,the blank group and the high-dose group were dissected,and the internal organs were collected to determine the organ coefficients,and the tissues were fixed and then pathological sections were made.Compared with the blank control group,there was no significant difference in piglets’body weight,body temperature,heart rate,respiratory rate and other clinical indicators in the low-dose,medium-dose and high-dose groups(p>0.05),indicating that the oral solution of MOS had no effect on piglets’body weight,body temperature,pulse,respiration and other clinical indicators.Compared with the blank control group,there was no significant difference in the blood routine and blood biochemical indexes of piglets in the low-dose,medium-dose and high-dose groups(p>0.05).It was found that there was no abnormal change in the shape,color and size of the organs of experimental piglets,and there was no significant difference in the organ coefficient between the drug group and the blank group(p>0.05).Compared with the blank group,the histopathological observation showed slight capillary hyperemia and inflammatory cell infiltration,and the organs all had normal structure.3.Observation on the effect of MOS on the cough and asthma symptoms of piglets artificially infected with infectious pleural pneumoniaFifty piglets of infectious pneumonia model were randomly divided into five groups,10 for each group,which were:the low-dose group(0.139ml/kg),the medium-dose group(0.278ml/kg),the high-dose group(0.556ml/kg),the positive control group(0.625ml/kg)and the model control group.The drug was administered by gavage once a day for 7d.Through clinical observation,pathological dissection observation,blood routine blood biochemical detection,viscera index observation,the symptom rate and mortality related indicators were calculated.The results showed that:during the experiment,there was no significant difference between each group before the administration according to the clinical symptom score(p>0.05).After 7 days of administration,the scores of each drug group were significantly reduced compared with those before administration(p<0.05),while there was no significant change in the model group(p>0.05).The score of the drug group was significantly lower than that of the model group(p<0.05).The symptom score of the high-dose group was lower than that of the positive control group.No piglets died in the high-dose and medium-dose group of MOS,the mortality rate was 20%in the low-dose group,30%in the positive group,and 50%in the model group.The body temperature and respiratory rate of the MOS in the treatment group were higher than those of the positive drug group and the model group.Among them,the body temperature and respiration rate of the high-dose group of MOS were 100%,the body temperature and respiration rate of the medium-dose group were 80%,and the respiration rate of the medium-dose group was70%.(3)effects of MOS on blood routine and blood biochemical indexes of piglets:leucocytes increased in the low,medium and high dose groups,while leucocyte counts decreased in the model group and the positive control group.The leucocyte counts increased significantly in the medium dose group compared with the other groups(p<0.05).The blood biochemical results showed that the content of alanine aminotransferase in the high-dose oral solution of MOS was significantly higher than that in the model group(p<0.05).The calcium content of the four drug groups was significantly higher than that of the model group(p<0.05).There was no significant difference in other indicators(p>0.05).In summary,the oral solution of MOS alleviated the cough response of capsaicin by reducing the expression of TRPV1 gene.The safety test of target animals showed that MOS was nontoxic to the target animals(pigs).MOS high dose to the pig infectious pleuropneumonia caused by cough symptom relief,MOS after the treatment,symptom scores decreased,sick pigs cough symptoms improved,explain to the pig MOS infectious pulmonary pleura pneumonia caused by good cough.
