| Barley(Hordeum vulgare.L)is one of important food crops in China,and the growth of barley is limited by phosphorus nutrition.Although there are various types and large proportions of soil organic phosphorus sources,the growth and development of barley are still limited.Therefore,it is of great significance to find out the suitable types and concentration of soil organic phosphorus for the growth of barley.At the same time,the genotypic difference of barley is also one of the important factors for the difference of soil organic phosphorus utilization ability.Therefore,this study selected the most suitable organic phosphorus source for wild type(CN4027)and cultivated type(Baudin)of barley growth by setting different types of organic phosphorus sources,and discussed the organic phosphorus utilization capacity of barley under different phosphorus concentrations.Based on the early QTL mapping of barley phosphorus utilization,the target genes related organophosphorus utilization were obtained by cloning,sequencing,and bioinformatics analyzing.The main results are as follows:(1)Barley can grow in an environment where Phytin,ATP,RNA,F6P and IHP organic phosphorus are the sole phosphorus sources.Under ATP and Phytin organic phosphorus treatments,the two varieties of barley can improve utilization ability of organic phosphorus by changing root morphology and increasing acid phosphatase and phytase activities,so as to get the biomass,phosphorus content and phosphorus accumulation equivalent to that of inorganic phosphorus treatment.At the same time,the phosphorus utilization efficiency of plants is related to their acid phosphatase activity,and the enhancement of phytase activity is beneficial to the accumulation of shoots biomass.(2)There was a genotype difference in the utilization capacity between the two varieties of barley under Phytin treatments.And the barley Baudin had a higher phosphorus utilization capacity of Phytin than CN4027.CN4027 had the best shoots dry weight at 3.75m mol·L-1 calcium phytate,and Baudin had the best shoots dry weight at 2.5 m mol·L-1.Barley enhances the utilization capacity of high-concentration Phytin by increasing root-to-shoot ratio,roots length,root surface area,root volume,and number of root tips,and by enhancing roots and secretory acid phosphatase activity.CN4027 mainly enhances the utilization ability of organic phosphorus by the enhancement of phytase activity,and Baudin enhances the utilization ability by the enhancement of acid phosphatase activity.(3)Genes MLOC_56200,MLOC_61737,MLOC_69490,MLOC_38965,and AK354580 were obtained by screening early QTL mapping of barley phosphorus.The results of gene cloning and gene relative expression show that the relative expression of these genes is affected by the concentration of organic phosphorus except MLOC_38965.Among them,the expression of gene MLOC_61737 and reference gene JF274705 vary greatly.And gene JF274705 is one of Hv PAP I-2 family members,and its protein is secretory protein.It is predicted that JF274705 will affect the secretion of acid phosphatase under phosphorus stress.MLOC_61737 is one of Hv PHT1 family members,and its protein is not secretory protein.It is speculated that MLOC_61737 plays a role in the process of phosphorus transport upward and chloroplast in barley. |
