Over recent years research on the existence of FGSCs in mouse ovaries has created interest among the biomedical scientists.Especially at the self-new and differentation.To address the question,we investigated the function of Hh signaling in FGSCs self-new and differentiation.Hh signaling is well conserved,which involves in germ cell development of Drosophila,zebrafish and male mammals.In this study,we applied the supplement of Hh signaling small molecules and lentiviral expression system to regulate the activity of Hh signaling,and we further explored the function of Hh signaling in FGSCs proliferation and folliculogenesis.Hh signaling could significantly promote FGSCs proliferation though activated Gli1 and Ccnd1 expression,which confirmed by histomorphometry,immunohistochemistry,western blot and q RT-PCR.The use of follicles 3D culture sets out to address the function of Hh signaling in FGSCs folliculogenesis.Our studies have demonstrated that the Hh signaling pathway participates in the folliculogenesis,which advanced our knowledge of folliculogenesis.In this study,we applied FGSC and follicles 3D culture to investigate the mechanisms of folliculogenesis.Moreover,we establish a platform for mechanical contraction of folliculogenesis,which provided a new angle of view in other fields of reproduction.
