| Diarrhea is one of the most common diseases in clinic,cause great economic losses with high incidence rate and high mortality rate.Pathogenic Escherichia coli cause diarrhea is the most common in clinic.In this study,we used E.coli O8to establish diarrhea model in mice,and studied the effect of Babu-7 effective ingredients compound,(BSEIC)on intestinal injury and immune function of diarrhea mice,in order to provide theoretical basis and experimental data for clinical treatment of diarrhea.85 Kunming SPF mice were divided into normal control group(NC group),negative group(UC group),ciprofloxacin group(CIP group)and BSEIC-H,BSEIC-M and BSEIC-L groups.Except for the NC group,the mice in each group were given 2%Na HCO30.3 m L/d by intragastric administration,afte 1×109CFU/m L E.coli O80.5m L were intragastric administration with mice.At the end of modeling,corresponding drug treatment was given for 3 days.The following tests were carried out:1.The diarrhea rate and diarrhea index of mice in each group were statistically analyzed.The results showed after E.coli O8intervention the diarrhea rate and diarrhea index in each group were no significant difference(P>0.05).The diarrhea rate and diarrhea index in BSEIC group were significantly lower than UC group(P<0.05)2.The changes of jejunum and ileum were observed by HE staining.The staining results showed that BSEIC could improve the length of villi,reduce the space between villi and alleviate the shedding of intestinal epithelial cells.BSEIC can repairing intestinal injury induced by E.coli O8in mice.3.The ultrastructure of ileum was observed by transmission electron microscope.The results showed that the invasion of E.coli O8resulted in short and disordered micro villi in the ileum of mice,and BSEIC could reverse the damage trend and make it normal.4.PAS staining was used to observe the number of goblet cells in ileum.The results showed that E.coli O8invasion reduced the number of ileal goblet cells in mice,and BSEIC intervention could effectively restore the decrease of ileal goblet cells caused by E.coli O8invasion.5.ELISA was used to detect IL-1,IFN-γ,TNF-α,IL-6,IL-4,IL-10,Ig G and IgA contents in serum and ileum s IgA.The results showed that compared with UC group,BSEIC could significantly reduce serum IL-1,IFN-γ,TNF-αand IL-6 content and significantly increased IL-4,IL-10,Ig G and IgA content(P<0.05).6.The results showed that BSEIC could significantly improve the content of CD3+,CD4+and CD19+cells in peripheral blood of mice compared with UC group,and significantly reduce the percentage of CD8+cells.Conclusion:BSEIC can repair the intestinal injury caused by E.coli O8,reduce the expression of proinflammatory factors in serum of infected mice,and improve the immune function of the body. |
