| Chinese medicine resources is the material basis for the development of traditional Chinese medicine industry,as on the international recognition and acceptance of traditional Chinese medicine,especially in this special time,significant achievements have been made in the process of resistance to disease of traditional Chinese medicine,provides reference for the world of Chinese,traditional Chinese medicine by increasing attention in the world,however,at present,the Chinese medicine resources are scarce.Salvia miltiorrhiza is a perennial herbaceous plant.Root rot is common in the cultivation process of Salvia miltiorrhiza.The incidence of serious plot is 60%~80%,which greatly reduces the yield and quality of Salvia miltiorrhiza and seriously affects the development of Salvia miltiorrhiza industry.Salvia miltiorrhiza root rot is a typical soilborne disease with the characteristics of rapid spread and difficult control.Currently,there are no means and methods for the control of Salvia miltiorrhiza root rot,but chemical methods are mainly used for the control.Chemical control has great influence on the quality of medicinal plants and may harm human health in clinic.Biological control is efficient,durable,green and pollution-free,so it is more suitable for the prevention and control of medicinal plant diseases.In order to investigate the resistance to catalyse of danshen just microorganisms to provide functional strains for salvia miltiorrhiza biological control,the experiment of salvia miltiorrhiza rhizosphere actinomycetes disease-resistant function to filter the growth,to evaluate and from its disease-resistant ability,both in biological control to provide effective biological danshen root rot disease resistant pay-off strains,and late for multifunctional microbial agents to provide theoretical basis for the development and utilization.The main research results are as follows:(1)Seven strains of rhizosphere actinomycetes of Salvia miltiorrhiza with strong bacteriostatic effect were screened.A total of 7strains were screened by plate confrontation test,and the average inhibition rate of the 7 strains against F.oxysporum and F.solani was more than 50%.(2)The rhizosphere actinomycetes of Salvia miltiorrhiza with phosphorus-soluble,iron-producing carriers and indole acetic acid production were screened and obtained.The phosphorus-dissolving activity of actinomycetes was detected by NBRIP petri dish detection method.A total of three strains of actinomycetes with phosphorus-dissolving activity were screened,and their numbers were SMRA-149,SMRA-150 and SMRA-160,respectively.By CAS plate screening method,39 strains of actinomycetes with ferro-producing vector activity were detected,and 10 strains with relatively strong ferro-producing vector activity.Salkowski colorimetry was used to detect the IAA production activity of the strains,and it was found that the IAA production capacity of the strains SMRA-100,SMRA-117 and SMRA-220 was 20.67 μg/mL,87.61 μg/mL and 30.40 μg/mL,respectively.(3)Identification of disease resistance of actinomycetes The pot experiment results showed that the rhizosphere release strain SMRA-220 of Salvia miltiorrhiza had a good control effect on root rot of Salvia miltiorrhiza,and the control effect increased by 43.82%.Compared with the Fo treatment group,the incidence of Fo + SK in the treatment group treated with actinomycetes decreased by 84.61%,the disease index decreased by 80.82%,and the control effect increased by 43.82%.The fresh weight and dry weight of the treatment group supplemented with actinomycetes increased by 138.45% and 65.88% compared with the Fo group supplemented with pathogenic bacteria.The root length,fresh root weight and dry root weight of the treatment group were significantly higher than those of the treatment group supplemented with pathogenic bacteria,indicating that the actinomycetes SMRA-220 had a good effect on the control of root rot of Salvia miltiorrhiza.(4)Evaluation of growth-promoting ability of actinomycetes Pot experiment results showed that the rhizosphere release strains SMRA-117,SMRA-220 and SMRA-100 significantly promoted the growth of Salvia miltiorrhiza seedlings.The treatment groups with smra-117,smra-220 and smra-100 spore suspension of actinomycetes significantly promoted the growth of salvia miltiorrhiza seedlings,and its fresh weight was significantly higher than that of the control group(P<0.05).Compared with the control group,the fresh weight of Salvia miltiorrhiza seedlings in SMRA-117 treatment group was increased by 234.64%,the fresh weight of root was increased by 226.08%,and the fresh weight of aboveground part was increased by 246.57%.Compared with the control group,the fresh weight of Salvia miltiorrhiza seedlings was increased by 85.23%,the fresh weight of roots was increased by 103.50%,and the fresh weight of aboveground part was increased by 59.79% in the SMRA-220 treatment group.In conclusion,the rhizosphere actinomycetes SMRA-117,SMRA-220 and SMRA-100 of Salvia miltiorrhiza can be used as the microbial source of growth promoting bacteria in the rhizosphere of Salvia miltiorrhiza.In conclusion,through screening the disease-resistance and growth-promoting function of rhizosphere actinomycetes of Salvia miltiorrhiza,and evaluating its disease-resistance and growth-promoting ability,a strain of rhizosphere actinomycetes with strong control effect on root rot of Salvia miltiorrhiza was screened.At the same time,three strains of rhizosphere actinomycetes of Salvia miltiorrhiza were screened,which had obvious promoting effect on the growth of Salvia miltiorrhiza seedlings.The results provided a research basis for the development and utilization of rhizosphere actinomycetes of Salvia miltiorrhiza and the green prevention and control of root rot. |
