In this paper, the cross-resistance risk to three neonicotinoid insecticides of Bemisia tabaci MEAM1 cryptic species was evaluated. The key genes of cytochrome P450 superfamily probably related to neonicotinoid cross-resistance were screened by transcriptiomic analysis and testified by real-time quantitative PCR.Relevant key gene functions were further verified by RNA interference to find out the key genes controlling the cross-resistance of MEAM1 cryptic species against the three neonicotinoid insecticides imidacloprid, acetamprid and thiamethoxam.Hopefully, the results will provide a theoretical basis for scientific control of Bemisia tabaci. The main content and research results are as follows:1.Threshold analysis method was used to study the resistance level and differences of MEAM1 cryptic species under long-term selective pressure of imidacloprid, thiamethoxam, and acetamiprid. Results showed that the risk of cross-resistance of imidacloprid-selected strain against thiamethoxam was less than the risk of cross-resistance to acetamiprid;the risk of cross-resistance of acetamiprid-selected strain against thiamethoxam was less than the risk of cross-resistance to imidacloprid, the risk of cross-resistance of thiamethoxam-selected strain against acetamiprid was less than the risk of cross-resistance to imdacloprid.2.Transcriptomic results were verified by real-time fluorescence quantitative PCR.In the thiamethoxam-selected strain of MEAM1 cryptic species, the cytochrome P450 9e2-like gene and the probable cytochrome P450 6a14 gene may be related to the cross-resistance of imidacloprid and thiamethoxam. While, probable cytochrome P450 305a1 gene and cytochrome P450 4g15-like transcript variant X2 gene may regulate cross-resistance between imidacloprid and acetamiprid of MEAM1 cryptic species, and cytochrome P450 4C1-like gene may regulate cross-resistance of MEAM1 Bemisia tabaci to the three agents, imidacloprid, acetamiprid, and thiamethoxam.3.RNA interference test showed that cytochrome P450 4C1-likel is a key gene related with the cross-resistance of Bemisia tabaci MEAM1 cryptic species against imidacloprid, thiamethoxam, and acetamiprid.
