The Verticillium Wilt in Cotton was caused by the Verticillium dahliae Kleb.It was fairly common in cotton production,and difficult to prevent and control.It was called_"the Cotton Cancer".This study choosed 26 kinds of cotton varieties(lines)for resistance identified indoors and in the field.The experiments were designed by chosen 4 kinds of seed coating agents and CQJ-5 to 3 and CQJ-5,investigated the disease index of Verticillium Wilt,selected the seed coating agents that had control effect on Verticillium Wilt.It searched the Verticillium Wilt StDAHP(GU479467.1,3-deoxy-D-arabino-heptulosonate 7-phosphate)in the GenBank.It searched 3 homologous genes in Gossypium hirsutum genetics database,cloned and named it,and bioinformatics analyzed.When cotton was induced by Verticillium dahliae.RT-PCR analyzed the target gene’s expression in leaves of resistant and susceptible varieties,Gossypium hirsutum L.varieties(lines)induced by Verticillium Wilt.The experiments aims to find the effective measures for control Verticillium Wilt of cotton.The main results were as follows:1.Resistance identified 21 kinds of varieties.Xinhai 13 was a disease resistant variety,relative disease index was 17.66 indoor and 15.31 in field.Suggested that the duration of indoor resistance identification should be extended to 60 days.2.Design experiments with 4 kinds of seed coating agents and CQJ-5 to 3,CQJ-5 by 10 treatments.Among these treatments,treatment 3 was the best(seed coating agent M2+CQJ-5 to 3).Its seed emergence rate was 86.56%.Its disease index was 25.83.Its control effect was 16.51%.Its lint yield was 20.83%higher than treatment 9(CQJ-5 to 3).3.3 target genes were cloned and named as GhDAHP-1,GhDAHP-2 and GhDAHP-3.It bioinformatics analyzed GhDAHP-1,its sequence length was 1983 bp,total amino acids was 539,molecular weight was about 59.45378 kDa,theoretical isoelectric point(pI)was 8.7.It bioinformatics analyzed GhDAHP-2,its sequence length was 1832 bp,total amino acids was 516,molecular weight was about 57.02998 kDa,theoretical isoelectric point(pI)was 7.68.It bioinformatics analyzed GhDAHP-3,its sequence length was 1652 bp,total amino acids was 5 1 7,molecular weight was about 56.95491 kDa,theoretical isoelectric point(pI)was 8.52.It did phylogenetic tree analysis,the relationship between GhDAHP and Gossypium raimindii with Gossypium arboreum and Gossypium arboreum were the closest.4.This study has sampled the cotton leaves after vaccinated by Verticillium dahliae for 0 hour,6 hours,12 hours,24 hours,48 hours,72 hours.It used ubiquitin protein 7 of island cotton(ubiquitin 7,GbUB7,DQ116441)as reference gene,and RT-PCR analyzed GhDAHP ’s expression in leaves.The results showed that after vaccinated by Verticillium dahliae,every varieties expressed differently at different points of time.There was a tendency to go up first and then down.Among the resistant varieties,the expression of GhDAHP was higher than the expression of susceptible varieties.Among the Gossypium hirsutum L.varieties lines,each lines had different expressions,the timing of reaching the peak was also inconsistent.It was speculated that after being induced by Verticillium dahliae,GhDAHP may be involved in the defense process of Verticillium Wilt infection,but has yet to be futher discussed in molecular aspects.It was still need to be researched about how does regulate the specific function of resistance to Verticillium Wilt in Gossypium hirsutum L.
