| Fat mass and obesity associated gene(FTO)ware involved in lipid metabolism and can regulate the body’s fat content through lipolysis.Leptin was secreted by fat cells,and it acts directly on the hypothalamus energy regulation centre to regulate the intake of body,thus playing a role in regulating body fat metabolism.The mechanisms and signaling pathways by which FTO and Leptin regulate fat production remain unclear.In this study,this experiment studied the relationship between FTO and Leptin expression levels and fat deposition in tail fat and abdominal subcutaneous adipose tissue of altay altay sheep of different ages under the conditions of diet with different body weights and energy levels,and to explore the relationship between FTO and Leptin and fat deposition.(1)The expression of FTO and Leptin and the regulation of fat deposition in the subcutaneous adipose tissue of the tail and abdomen of altay sheep at different ages mensuelsBy daily weight gain,24 male Altay sheep with the fastest growth rate(the high-weight group,weight was in the top 10%)and 24 male altay big-tail sheep with the slowest growth rate(the low-weight group,weight was in the bottom 10%)were selected as experimental subjects.Six were in the low-weight group and six were in the high-weight group at 3 months of age,the rest are fed to 6 months of age,six of the low-weight and high-body reorganizations were slaughtered,the remaining experimental animals were used for the next experiment.Weigh before and after slaughter.Weighing before and after slaughter,blood,tail fat and abdominal subcutaneous fat were collected.The size of adipocytes was determined by HE staining,the area of adipocytes was determined,and the gene and protein expressions of FTO and Leptin in adipose tissue were determined by real-time fluorescence quantitative PCR and Western blotting.The results showed:The contents of T3,T4,FABP4,FASN,TSH,GAPD and Leptin as well as subcutaneous adipose tissue in the tail and abdomen of the high-weight altay sheep group at 3 months and 6 months were significantly higher than those of the low-weight group(P<0.05),the area of fat cells in the high-weight group at 3 months and 6 months was significantly higher than that in the low-weight group(P<0.01),the expression of FTO was significantly lower than that in the low-weight group(P<0.05),and the expression of Leptin was significantly higher than that in the low-weight group(P<0.05).The expression level of FTO in the low-weight group at 3 months and 6 months was significantly higher than that in the high-weight group(P<0.05),and the expression level of Leptin in the low-weight group at 3 months and 6 months was significantly lower than that in the high-weight group(P<0.05).The results showed that:Under the same feeding condition,the expression of FTO was low and Leptin was high in the high-weight group and the expression of FTO was high and Leptin was low in the low-weight group in the low-weight group of altay sheep.(2)Effects of diets with different energy levels on FTO and Leptin expression in subcutaneous adipose tissue of the altay sheep tail and abdomenThe control group was fed a diet designed according to the standard(2004),in which the energy intake was 100%of the nutritional requirement.The 6-month-old high-weight group was divided into the control group and the low-energy group,the low-weight group was divided into the control group and the high-energy group.After feeding for 30 days in groups,weigh before and after slaughter,and the blood,tail fat and abdominal subcutaneous fat were collected for the purpose of studying the blood biochemical indexes,fat morphological changes and FTO and Leptin expression levels of sheep.The results showed:Altay sheep of low weight group were fed with high energy feed,the contents of T3,ADP,GAPD,Leptin and adipose tissue in serum of the high-energy group were significantly higher than those of the control group(P<0.05),the area of fat cells was significantly higher than that of the control group(P<0.05),expression levels of FTO gene and protein were significantly lower than those in the control group(P<0.05),leptin expression was significantly lower than that of the control group(P<0.05).Altay sheep of high weight group were fed with low energy feed,the contents of T3,T4,TSH,S.S,PG,ADP,GAPD,FABP4,Leptin and adipose tissue in the serum of the low-energy group were significantly lower than those of the control group(P<0.05),the area of fat cells in the low-energy group was significantly lower than that in the control group(P<0.05),the expression of FTO was significantly higher than that of the control group(P<0.05),leptin expression was significantly lower than that of the control group(P<0.05).The results showed that:Under the conditions of diet with different energy levels,the body weight and body fat of the high-energy group in the low-weight group increased significantly,and the expression of FTO showed a downward trend,while the expression of Leptin showed an upward trend.However,the body weight and body fat of the low-energy group in the high-weight group decreased,and the expression of FTO increased,while the expression of Leptin decreased. |
