As a main ornamental plant,rose is delightful due to its bright color,full aroma and graceful shape,which is often used as medicine and tea,and endowed with unique cultural connotation.Rose has long been planted in China,and currently cuttage is believed to be the main cultivation method which is also widely applied to industrialized production.However,there is a shortage of comprehensive evaluation methods to assess the rooting ability of various roses.Learning rooting characteristics of rose is conducive to seedling production.Besides,it is essential to make breakthrough in new substances on the basis of traditional rooting agent in a bid to further improve survival and rooting rate.Firstly,this study preliminarily explored the cutting propagation technology of rose.Based on this,it assesses the rooting ability of diverse roses,and discusses impacts of different exogenous substances on the rooting of rose.The results are as follows:1.Rose is a mixed rooting type.The root primordium is induced both callus and cambium cell touching with the pith ray.Besides,the rooting rate is determined by the reserved lobules number of cuttings and planting method.Compared with cuttings without lobules,the cuttings reserved 2-5 lobules is likely to grow better.And compared with hydroponics,the solid medium culture is more efficient in terms of rooting and survival.2.After 20 days of cuttage,four root indexes of 50 kinds of roses were measured,including average rooting rate,average root length,average number of root and longest root.On the basis of the PCA(principal component analysis)of the four indexes,the comprehensive value(Z value)of different varieties was calculated.It is concluded that there is a significant difference among different varieties.The results of comprehensive evaluation values showed that the Rosa.’Accademia’ rooting ability is the most outstanding.Most varieties’ rooting ability is moderate,but Rosa.’Angela’,’KONGQUEKAIPING’,’Tancho’,’Unconditional love’ and ’The Impressionist’ are worst for they didn’t grow roots within 20 days.Taking clustering map and comprehensive index into account,the rooting ability of rose is graded as four levels:strong,good,mediocre and poor.The first one only accounts for 18%.Most varieties have yet to be improved.3.In the research,four substances that could effectively enhance rose’s rooting are picked.The study reveals that MeJA,SNP,H2O2 and glucose have positive effects on rose’s cuttage.Rosa.’Old Blush’ that has medium capability of rooting was used as experimental material and four kinds of exogenous substances’ concentration gradient experiment was conducted.The result showed exogenous substances’ application for 30min in the proper concentration before inserting the matrix has promoting effects.Comparing the effects of different substances at an optimal concentration,the effect of MeJA is best,glucose is second,and H2O2 and SNP are a bit poor.The optimal concentration of MeJA,glucose,H2O2 and SNP is advised to be 0.25 mg/L,2.0 g/L,0.10%and 0.75 g/L respectively.4.The research studied the cooperative effect of glucose,H2O2 and SNP,whether they can further promote the root regeneration by the common application.The result shows that three combination treatments(H2O2+glucose,H2O2+SNP,H2O2+ SNP+glucose)could be better than single use of any substance.If H2O2,glucose and SNP are used together,the effect will be best.
