| Diabetes mellitus(Diabetes)is a metabolic disease caused by the deficiency of insulin in the pancreas in the body due to the long-term high fat diet.Nonalcoholic Fatty liver disease(NAFLD)is a common pathological lesions in the liver,caused by a variety of conditions rather than a single metabolic disease.At present,in addition to exploring effective drug therapy,the research on prevention of diabetes and fatty liver by using natural plant has been paid more and more attention.The acacia honey is made by honey bee who collects the pollens from acacia flower.The previous researches have found that acacia honey reduces blood pressure effectively,improves blood circulation,prevents the blood vessel stiffness and inhibites inflammation.Previous researches for healthy beneficial effects of bee honey mainly focus on improving gastrointestinal function,promoting digestion and absorption,improving the immune response.The research on its hypoglycemic function has not been reported yet.The present study is to explore the effects and mechanisms of different concentrations of acacia honey(1g/kg,2g/kg,3g/kg)on fatty liver formation and hypoglycemia in C57BL/6 mice induced by high lipids,.The main results obtained are as follows:1.In this animal experiment,C57BL/6 mice were fed with standard chow diet with free diet and drinking water.Adapt to the spf-grade animal house for a week and conduct experiments.After one week adaption,the animals were gavaged the different concentrations of acacia honey(1g/kg,2g/kg,3g/kg)continuously for 23 weeks.After 4weeks of bee honey treatment,there was no significant difference in body weight between various intervention.After 12 weeks of intervention,there was no significant difference in liver weight between the six groups of mice with bee honey intervention compared with those in the high-fat group.However,after 23 weeks of bee honey intervention,the liver weight in the high-fat diet group was significantly higher than that in the control group.However,The hepatic weight to body weight ratio in the high fat diet groups treated with low and medium dosage of bee honey were significantly lower than that in the high-fat group.2.In the prevention experiment,the blood glucose in the high-fat group was significantly higher than that in the low-fat group after four-week high fat diet feed.After 8weeks of the intervention,the blood glucose in the high-fat feed mice treated with low and medium dosage of bee honey were significantly lower than that in the high-fat group.And this hypoglycemic effect of low and medium dosage of bee honey intervention remains effectively until 23 weeks of bee honey intervention3.The pro-inflammatory cytokines in the liver were detected by Real Time PCR.The expression levels of IL-6,TNF α,MCP1,IL-1 beta were significantly inhibited by low and medium dosage(1 g/kg,2 g/kg)of bee honey intervention.The liver SREPB1 gene expression was also significantly inhibited.By Western blotting,the expression of GLUT2 protein was significantly increased with the treatment of 1g/kg BW bee honey at the 23 weeks of intervention,4.Hematoxylin and Eosin Staining of liver sections showed that low and medium dosage(1 g/kg,2 g/kg)of bee honey intervention significantly inhibited lipid accumulation in the liver cells,and prevented fatty liver formation of C57BL/6 mice induced by high fat diet. |
