| At present,with the development of economy and the improvement of living standard,obesity and related diseases caused byexcessive high glycolipid diet are becoming more and more serious.The side effects of clinical anti-obesity drugs can not be ignored,not suitable for long-time use.Therefore,dietary intervention is a safer method,and the development of functional foods is particularly important.This experiment mainly mainly studied the physicochemical properties of different resistant starches and modifiedstarch starches,and the effects of weight loss and lipid lowering on obesity induced by high lipid diet.Firstly,different types of modified starch were prepared from high amylose corn starch by pregelatinization and citric esterification.The results showed that the esterification degree of esterified starch was the highest at 150℃,and the esterification degree of starch could be further improved by pre-gelatinization treatment,which make it up to 0.33.Then the structure and physicochemical properties of the sample were determined.By measuring the in vitro digestibility,thermal stability and structural changes of the four samples,we can see that the physicochemical properties of the pregelatinized sample changed little,although the structure of the pregelatinized samples changed.After esterification,the two samples were cross-linked with citric acid due to the presence ofcitric acid on the surface,which not only changed its structure,but also decreased its digestibility,thus,it was more suitable for the treatment of obesity induced by high fat diet.Then,four kinds of starch were added to the feed of rat induced by high glucolipid diet for 8 weeks.60 rats were divided into six groups:normal group,high l ipid group,original starch group,pregelatinized starch group,esterified starch group and esterified pregelatinized starch group.The feeding amount,weight and blood sugar of rats were measured during the feeding period.The body fat,organ weight,serum insulin and blood lipid levels,liver function related indexes and pathological sections of fat and liver were observed.The results showed that the weight,fat weight,liver weight and body fat ratio of rats induced by high fat diet were decreasedin varying degrees.And the serum insulin level was reduce and the insulin sensitivity.was improved.The levels of TG,TC and LDL in serum of rats were decreased,and the levels of fatty acid metabolism and phospholipid metabolism were increased.The activity of transaminase and alkaline phosphatase in serum of rats were decreased,and cell steatosis in adipose tissue and liver tissue was inhibited.In particular,the intervention effect of esterified starch and esterified pregelatinized starch is more obvious.Finally,the expression levels of glucose and lipid metabolism related genes in fat and liver of rats were detected by RT-PCR.By analyzing glucose and lipid metabolism related genes(LXRα、HMGCR、Aocx1、GS2、GYG、Insig1、Insig2、SERBP、FATP1、Adiponectin、GLUT4、FABP4),it was found that the intervention can regulate the synthesis of fat and the conversion of glycogenin the body by regulating glucose and lipid metabolism genes,thereby playing a role in weight loss and lipid lowering.Conclusion:Finally,through the above in vitro and in vivo studies it is canbe found that ater chemical modification,citric acid esterification has the most obvious effect of reducing fat and reducing weight.,and after pretreatment of citric acid esterification sample effect is more superior.Therefore,this study provides a theoretical basis for the further effective development of new functional foods,and also provides a new idea for the prevention and treatment of metabolic syndrome caused by diet-induced obesity. |
