The Effect Of Key Protein Interferon Gene In CGAS-STING Signaling Pathway On Autophagy And Apoptosis In Human Bronchial Epithelial Cells

Objective: To study the effect of key protein interferon gene stimulating protein（STING）in cGAS-STING signaling pathway on autophagy and apoptosis of human bronchial epithelial cells,and provide new ideas for the treatment of chronic obstructive pulmonary disease.Methods :The STING gene silenced in beas-2b cells was compared with the normal group,and the cells were infected with adenovirus with green fluorescence（Ad V-GFP）.The m RNA expression of STING and β-IFN were detected by reverse transcriptionpolymerase chain reaction（RT-PCR）.The expression of STING,P62,LC3 B and caspase-3 in cells was detected by western blot.Expression of β-IFN was detected by enzyme-linked immunosorbent assay（ELISA）.Cell apoptosis was detected by flow cytometry.Results:（1）STING molecules are expressed in BEAS-2B cells,and after STING gene silencing,the expression of β-IFN is down-regulated（p<0.01）,the expression of autophagy related protein P62 is upregulated（p<0.01）,and the ratio of LC3B-II/LC3B-I is down-regulated（p<0.01）.（2）the expression of STING was up-regulated after infection of human bronchial epithelial cells（p<0.01）.After the infection of Adv with beas-2b,the expression of autophagy related protein P62 was up-regulated（p<0.01）,the ratio of lc3b-ii/lc3b-i was decreased（p<0.01）,and the expression of apoptotic protein active caspase-3 was decreased（p<0.01）.Conclusion: adenovirus infection can induce enhanced autophagy in human bronchial epithelial cells,and adenovirus combined with CSE infection can synergistic enhance autophagy.The silence STING gene can inhibit autophagy in human bronchial epithelial cells;The cGAS/STING/IFN signaling pathway may play a role in the autophagy and apoptosis of human bronchial epithelial cells induced by adenovirus infection.
