ObjectiveTo investigate the effect of whole body vibration on the autophagy activity and energy metabolism of gastrocnemius in type 2 diabetic mice.MethodsMice were randomly divided into three groups: Nomal control group(normal diet,n=12),Diabetes control group(high-fat diet+STZ,n = 12),and Diabetes vibration exercise group(high-fat diet+STZ,n = 12).After fed with high-fat diet,injected with STZ and blood sugar reaches 11.1 mmol/L,the basic physiological indicators using weight,blood glucose,and lipid measurement were observed at2,4,6,8,10,and 12 weeks after the whole body vibration intervention.The expression changes of autophagy marker proteins(Beclin-1 and Atg7)and energy metabolism indicators(G6P,SREBP-1 and FAS)were detected by Western blot and RT-q PCR,respectively.HE staining was used to observe the morphological changes of skeletal muscle of diabetic mice.ResultsBody weight results showed that the weight of type 2 diabetic mice was significantly heavier than that of normal mice,while there was no significant difference in weight between the control and whole body vibration groups.Bloodglucose results showed a significant decrease in blood glucose levels after 12 weeks of whole body vibration treatment.Blood lipid results showed that whole body vibration had no effect on TC,TG concentrations in mice,and was a significant difference in LDL and HDL levels.On the 12 weeks after high-fat diet,western blot and RT-Qpcr showed that the expression of autophagy marker proteins Beclin-1 and Atg7 were significantly up-regulated by whole body vibration,and the expression of glucose metabolism-related protein G6 P was significantly enhanced,while lipid metabolism was significantly inhibited.Moreover,HE staining showed that whole body vibration significantly increased the skeletal muscle muscle area of the gastrocnemius muscle in type 2 diabetic mice.ConclusionsWhole body vibration markedly activates autophagy activity and energy metabolism of gastrocnemius muscle in type 2 diabetic mice,and has neuroprotective effect.
