| Objective: To investigate the relevance between Beclin-1、LC3 in PBMCs of newonset IgAN with clinical symptoms、experimental biochemical data and renal pathological data.Methods: Beclin-1、microtubule-associated protein 1 light chain 3(LC3)mRNA in PBMCs of the IgAN group(n=30)and healthy control group(n=30)were tested by real-time fluorescent quantitative PCR(RT-PCR).Beclin-1、LC3 proteins in peripheral blood mononuclear cells of IgAN group and the healthy control group were tested by immunohistochemical method;and Beclin-1、LC3 proteins in the kidney tissues of IgAN group were tested by immunohistochemistry of frozen sections.To investigate the relationship between clinical data and Beclin-1、LC3mRNA.Results: 1.LC3 mRNA in peripheral blood mononuclear cells of IgAN group(n=30)was lower than that in the healthy control group(n=30)(P < 0.001),while Beclin-1 mRNA was not statistical difference between IgAN group with the healthy control group(P> 0.05).2.Compared with healthy controls,the expression rate of LC3 protein in peripheral blood mononuclear cell of patient with new-onset IgAN was lower,and Beclin-1 was higher.Meanwhile,Beclin-1proteins and LC3 proteins could be expressed in the kidney tissues of patients with new-onset IgAN.3.In IgAN group,comparison between the Beclin-1mRNA low-level group(n=15)and the Beclin-1 mRNA high-level group(n=15),the eGFR,HCO3-was significantly lower and urea,uric acid was significant difference higher in Beclin-1 mRNA high-level group(P<0.05),however,there was no significant difference of the two groups in other laboratory data,clinical symptoms and renal pathological data(P>0.05).4.In IgAN group,there was not statistical difference of clinical symptoms,laboratory data and renal pathological data between the LC3 mRNA high-level group(n = 15)and the LC3 mRNA low-level group(n = 15)(P>0.05).Conclusions: 1.The down-regulation of LC3 mRNA in peripheral blood mononuclear cells of patients with new-onset IgAN may be involved in the development of IgAN by influencing autophagy activities.2.The up-level of Beclin-1 mRNA may affect eGFR and tubular reabsorption of HCO3-. |
