Research backgroundWith the development of people’s living standards,their diet structure was changed greatly.Diabetes mellitus is one of the metabolic diseases,its incidence rate is increasing year by year.Most of the complications of diabetes are chronic complications,mainly vascular and neuropathy.In particular,diabetic foot ulcer is in chronic inflammatory state for a long time,which leads to the wound prolongation.Most of the complications of diabetes are chronic complications,mainly vascular and neuropathy,while diabetic foot ulcer is in chronic inflammatory state for a long time,which leads to the wound prolongation.Macrophages can affect the secreted inflammatory cytokines in different polarization states.Cytokines such as VEGF and TNF-α play a key role in the wound healing process.Autophagy is a transport pathway of cell membrane,which has a potential connection with inflammatory wound repair.At present,the research on the pathogenesis of diabetic chronic wound is increasingly in-depth at home and abroad.It has been found that autophagy participated in the regulation of macrophage polarization,which can cause different polarization of macrophages and affect the process of chronic inflammatory disease.At the same time,some studies have shown that the wound healing of diabetes mellitus is closely related to the polarization of macrophages.PurposeTo study the effect of human intervention on autophagy on the polarization of macrophages and the healing of chronic inflammatory wounds in diabetes.MethodsTo study the effect of autophagy and macrophage polarization on diabetic chronic inflammatory wound healing.RAW264.7 macrophages were cultured by high glucose culture method to construct a diabetes mellitus cell model.The cell proliferation effect of RAW264.7 in high glucose culture medium was detected using CCK-8 Assay to select the appropriate glucose concentration to build a diabetes mellitus cell model.The experiment was divided into inducer group,inhibitor group,and blank control group,and the detection time nodes sets 6H,12H,18H,24H,and 48H.the cell model of diabetes mellitus was constructed.RAW264.7 cells were co-cultured with resveratrol,3-MA and high glucose culture medium with additional glucose,respectively.The protein expression differences of macrophage polarization markers CD16(M1),CD206(M2),F4/80(MO)and autophagy related p62 and LC3 were detected using western blot and immunofluorescence.The mRNA expression level of TNF-α,IL-4,VEGF and IL-6 in diabetic chronic inflammatory wound healing was detected using qPCR.ResultsThe cell proliferation ability of RAW264.7 macrophages in different concentrations of high glucose medium was detected using CCK-8,and a cell model of type 1 diabetes was constructed.The results of western blot and immunofluorescence showed that the expression level of LC3 and CD16 was significantly increased while the expression of p62 was significantly decreased in the resveratrol group.And the expression level of LC3 in the 3-MA group was significantly reduced while the expression of P62 increased,and the expression of CD206 on the surface of M2 macrophages increased.The mRNA expression level of TNF-α,IL-4,VEGF and IL-6 in all detected groups indicated that autophagy could regulate the polarization of macrophages and regulate the wound healing of diabetic chronic inflammation.Conclusion1.Resveratrol can induce the macrophage autophagy and induce macrophage to polarize into M1 macrophage.2.3-MA can simulate macrophages to polarize into M2 macrophages through inhibiting macrophage autophagy.3.Regulating autophagy can affect the transformation of macrophages to different polar states and affect the polarization macrophages to secrete TNF-α、IL-4 and VEGF、IL-6 inflammatory factors,thereby regulating the inflammatory response in the wound,and affect the healing of chronic inflammatory wound in the chronic inflammatory wound of diabetes mellitus.
