Objective:To investigate the effects of adipose-derived mesenchymal stem cells on asthma and ILC2s in asthmatic models.Methods:Adipose-derived mesenchymal stem cells were isolated from male C57BL/6 mouse and then identified.40 wild-type C57BL/6 mice were randomly divided into the groups of blank control,asthma model,cell control and cell therapy groups.Asthma model were established by ovalbumin and then identified.Cell control and cell therapy groups were intravenously administered 1×10~5 ADSCs 24h after the last sensitization.24h after the last challenge,the lung tissue was taken for HE staining.And the distribution of GFP~+-ADSCs in lung tissue was observed.The BALF was used to count the exfoliated cells,then OVA-Ig E expression in the serum and BALF supernatant were detected to analyze the degree of airway inflammation.The lung tissue was digested into cell suspension and the ratio of ILC2s of each group was detected by flow cytometry.The expression of IL-5,IL-13,GATA3,ST2 and ICOS-L in lung tissue was detected by q RT-PCR to observe the effect of ADSCs transplantation on airway inflammation in asthmatic mice and its effect on ILC2s.Results:GFP~+-ADSCs were able to launch the lung tissue through the tail vein injection.In the asthma model group,there were obvious inflammatory cells infiltration around the bronchi and peripheral lung tissues.Airway inflammation was alleviated in the cell therapy group.The expression of OVA-Ig E in serum and BALF supernatant was higher in the asthma model group than in the control groups,and the cell therapy group was significantly lower than the asthma model group.Compared to the two control groups,the proportion of ILC2s in the lung tissue of asthmatsic mice was significantly higher,and lower in the cell therapy group;Moreover,the expression of IL-5,IL-13 in asthma group were up-regulated while those gene in cell therapy were down-regulated;the expression of GATA3,ST2 and ICOS-L in the asthma model group was significantly up-regulated,while the cell treatment group was significantly decreased.Conclusion:Activation of ILC2s is an important pathogenesis of bronchial asthma.ADSCs can inhibit airway inflammation,reduce the proliferation of ILC2s,impact the function of ILC2s,thus ADSCs can play a role in the treatment of bronchial asthma.
