| ObjectiveTo explore the value of detecting HBV DNA content in occulting hepatitis B virus infection(OBI)and the mechanism of OBI in serum with at least one positive surface antibody(HBs Ab),E antibody(HBe Ab)and core antibody(HBc Ab)negative hepatitis B surface antigen(HBs Ag)and hepatitis B e antigen(HBe Ag).MethodsThe serum markers of hepatitis B virus(HBV-M)were detected by chemiluminescent immunoassay(CLIA).The HBV-DNA content was re screened by real-time fluorescence quantitative polymerase chain reaction(PCR),and the detection of OBI was analyzed.Alanine aminotransferase(ALT),aspartate aminotransferase(AST)and total bilirubin(bilt3)were detected by wet chemical method to analyze the liver injury.Hepatitis C virus antibody(HCV-Ab),syphilis antibody(TP-Ab)and human acquired immunodeficiency virus antibody(HIV-Ab)were detected by CLIA method to analyze whether they were infected with other viruses and whether there were significant differences in host immune response.Sequencing method was used to detect whether HBV gene mutation existed and analyze the mechanism of OBI.ResultsDuring the experiment of this paper,1138 sera negative for HBs Ag and HBe Ag,with at least one positive for HBs Ab,HBe Ab and HBc Ab were collected.A total of 35 sera positive for HBV-DNA were detected by PCR,and OBI detection rate was 3.08%.The detection rate of HBc Ab,HBs Ab and HBc Ab positive HBV-M mode was the highest, which was 4.11% and 4.04%,respectively.The positive detection rate of HBV-DNA was significantly increased with the increase of the sample absorbance/cut-off value(S/ CO value)(P <0.05).596 patients received liver function examination, liver injury accounted for22.99%,liver injury group HBV-DNA positive rate,HBc Ab high reactivity(S/ CO≥10.0)were significantly higher than the normal liver group(P < 0.05).There was no significant difference in the group infected with other viruses(P >0.05).HBV gene mutation results to be the third party testing company reply.ConclusionsRoutine detection of HBs Ag and HBe Ag negative accompanied by HBc Ab positive indicated the possibility of OBI,and the risk of OBI increased significantly with the increase of S/ CO value of HBc Ab reactivity. At the same time, with reference to liver injury indexes,it is suggested that further detection of HBV DNA content should be performed to provide reference for clinical diagnosis of OBI and reducing the risk of HBV transmission caused by OBI. |
