Objective: The expression of CALHM1,ALKBH5,XIST and the enrichment of CALHM1 M6 a in the peripheral blood of diabetic patients were determined to analyze the correlation between the demethylation of CALHM1 and CALHM1 m6 A and diabetic neuralgia(DNP).To investigate the effect of XIST and ALKBH5 on the expression of CALHM1 on the demethylation of CALHM1 M6 a,and to provide clinical evidence for the clear role of CALHM1 M6 a demethylation in the pathogenesis of DNP..Methods: The subjects were patients admitted to the inpatient department of the First Affiliated Hospital and the Second Affiliated Hospital of the University of South China from March 2019 to March2020.The subjects were divided into three groups: normal control group(N group,N =15)was adults without diabetes;Control group(DM group,n=20)consisted of adults with diabetes but without DNP;The experimental group(DNP group,n=22)consisted of adults with DNP.Peripheric blood samples of the three groups were collected to detect the expression of CALHM1 M6 A demethylation,CALHM1,ALKBH5 and XIST in the peripheral blood of the three groups,respectively.To compare whether there were differences in its expression levels in the peripheral blood of the three groups;To analyze whether the duration of disease and Hb A1 c in DNP group were correlated with the expression of related indexes in peripheral blood.The expression levels of CALHM1 and CALHM1 M6 a methylation in peripheral blood of the three groups were compared,and their correlation with the course of disease and Hb A1 c of the patients was compared.The relationship between the M6 a methylation level of CALHM1 and the expression of CALHM1 was analyzed statistically.The effects of ALKBH5 and XIST expression on CAHM1 M6 a methylation level in peripheral blood of DNP group were analyzed statistically.The effects of ALKBH5 and XIST expression on CAHM1 expression in peripheral blood of DNP group were analyzed statistically.Results: 1.There was no significant difference in the basic data of the three groups(> 0.05).There were differences in disease course and(49)Hb A1 c between DM and DNP groups,with statistical significance(<(49)0.05).2.The expression of CALHM1 in peripheral blood of DNP group was higher than that of N group and DM group;With the progression of neuropathy and the increase of Hb A1 c content,the expression level of CALHM1 in peripheral blood of DNP patients increased.3.Compared with N group and DM group,the expression of CALHM1 M6 A methylation in peripheral blood of DNP group was lower;In DNP group,the expression of CALHM1 in peripheral blood increased with the progression of disease and the increase of Hb A1 c.4.In DNP group,with the decrease of CALHM M6 A methylation level,the expression level of CALHM1 in peripheral blood of patients was higher.5.There were differences in the expression levels of XIST and ALKBH5 in peripheral blood of the three groups,and the differences were statistically significant.6.In the peripheral blood of DNP patients,with the decrease of XIST,the methylation level of CALHM1 M6 a decreased,and the expression level of CALHM1 increased;With the increase of ALKBH5,the M6 a methylation level of CALHM1 decreased,and the expression level of CALHM1 increased.Conclusion: The high expression of CALHM1 in DNP patients is related to the demethylation of CALHM1 M6 a.The expression of XIST and ALKBH5 was correlated in DNP patients,and the latter two could affect the demethylation of CALHM1 M6 a in DNP patients.
