Background: Transcription factors specifically bind to the upstream specific sequence of the 5 ′ end of the target gene,so as to ensure the expression of the target gene at a specific intensity at a specific time and space.This study aims to prove that SP2 is the upstream transcription factor of Ski gene,which has specific binding sites and guides Ski to play a regulatory role in astrocyte proliferation.Methods: Using Jaspar and USCS databases to predict grafting and gradually reduce the threshold to screen the transcription factors upstream of Ski gene,so as to preliminarily determine the transcription factor SP2.Next,we analyzed the correlation between the expression of SP2 and Ski genes in normal astrocytes and reactive astrocytes,and the changes of astrocyte proliferation.In order to further confirm the effect of SP2 regulating Ski on astrocyte proliferation,si RNA targeting SP2 was transfected into astrocytes,and then the expressions of Ski,PCNA,Cyclin D1 and astrocyte proliferation were analyzed to evaluate the effect of SP2 knockdown on Ski and astrocyte proliferation.Finally,double luciferase reporter assay confirmed that transcription factor SP2 had a specific binding site in the promoter region of Ski gene.Results: According to the database analysis,SP2 was predicted to be the upstream transcription factor of Ski gene.SP2 had a specific binding site in the promoter region of Ski gene.There was a positive correlation between SP2 and the expression of Ski in astrocytes.Knockdown of SP2 could down regulate the expression of Ski gene and inhibit the proliferation of astrocytes.Conclusion: It is the first time to find and confirm that transcription factor SP2 plays an important role in the proliferation of astrocytes by targeting ski gene.This finding may help to find a new target for the treatment of spinal cord injury.
