Abstract/Summary:
Objective:1.To study the expression of PSAT1 in renal clear cell carcinoma.2.Study on the concentration-dependent effect of PSAT1 on the proliferation of renal cancer cells and its mechanism.Methods:1.Study on the expression of PSAT1 in cancer tissues by immunohistochemical experiment.2.Collecting cases to study the relationship between PSAT1 and clinical data.3.The effect of PSAT1 concentration on cells was studied by CCK8,Edu and colony formation assay.4.Study on the effect of PSAT1 concentration on cell proliferation by flow cytometry.Results:1.PSAT1 was highly expressed in renal clear cell carcinoma.And it is related to TNM stage and Fuhrman grade.2.PSAT1 promotes the proliferation of renal cancer cells in a concentration-dependent manner.3.PSAT1 could promote cell proliferation and clone formation,and Edu assay showed that the number and intensity of green fluorescence in the nucleus increased significantly after the treatment of PSAT1,that is it significantly promoted the proliferation of renal cancer cells.4.Flow cytometry showed that the proportion of early and late apoptosis in the PSAT1 group was significantly lower than that in the control group.Conclusion:PSAT1 is highly expressed in renal clear cell carcinoma.When PSAT1 acts on786 Mel O and ACHN cells,the proliferation and invasion ability of cancer cells are increased,and the apoptosis rate of cancer cells is decreased.
