| Objective:(1)The expression profile of urinary exosomal miRNA in IMN patients was detected comprehensively and systematically,and the differential miRNAs were found.(2)The specific differential miRNAs were verified by real-time quantitative polymerase chain reaction(RT-q PCR).(3)Receiver operating characteristic(ROC)curves and serum specific antibody,laboratory tests were used to evaluate the diagnostic value of exosomal miRNAs in IMN patients Methods:Urine samples were collected from 36 IMN patients and 36 healthy persons.Among them,6 cases of IMN and 6 cases of healthy subjects were used to analyze the miRNA profiles of urinary exosomes by high-throughput sequencing.Then the differential miRNAs screened by remaining 30 cases of IMN and the healthy persons were verified by RT-q PCR.Additionally,the serum of the 30 patients and healthy people were collected,and the clinical parameters were detected.Pearson,Spearman correlation coefficients and ROC curves were used to evaluate the diagnostic value of exosomal miRNAs in IMN patients.Results:The urinary exosomes were successfully separated and the exosomal miRNAs were obtained.Compared with the healthy control group,the expression levels of 20 miRNAs were down-regulated and 5 miRNAs were up-regulated in the urine exosome miRNA of IMN patients.From the verification cohort,the expression levels of miR-9-5p and miR-30b-5p were decreased in IMN patients,which was consistent with the results of high-throughput sequencing.Among them,miR-9-5p was correlated with e GFR and triglyceride.Mi R-30b-5p was related to anti-PLA2 R,ALB and β2-MG.The analysis of ROC curves revealed that the expression of miR-30b-5p and miR-9-5p showed a potential diagnostic value for IMN.Conclusion:This study showed that there were significant differences in urinary exosomal miRNA profiles between IMN patients and healthy controls.Mi R-9-5p and miR-30b-5p may become new non-invasive biomarkers of IMN. |
