Objective: For acute ischemic stroke patients with dyslipidemia with new card dialectical,statistical analysis of the syndrome factor distribution,record the blood lipid of patients,and the patients with phlegm wet syndrome factor for genetic testing,analysis of Apolipoprotein E gene rs429358,rs7412 and rs7259620 three Single nucleotide polymorphisms(SNP)site on a phlegmy wet the correlation of patients with acute ischemic stroke with dyslipidemia.Method: Filtered in hospitalized patients,in accord with stroke diagnostic criteria of traditional Chinese medicine and western medicine diagnostic criteria of acute ischemic stroke patients,through the electronic medical record system refer to patients after admission to collect a full set of indicators of blood fat,in patients with dyslipidemia diagnosis standard,by listening,asking and diagnosis of traditional Chinese medicine,professor wen-feng zhu initiative dialectical syndromes elements using this new system,the reference "syndrome factor differentiation to learn the theory,according to the ischemic stroke syndrome factors diagnosis scale of diagnostic criteria,of acute ischemic stroke patients with dyslipidemia of the dialectic,and carries on the statistical description of the syndrome factor distribution,Screening of patients with acute ischemic stroke with dyslipidemia in patients with phlegm wet dialectical elements,blood,from its early in the morning on an empty stomach with Bio Teke whole blood genomic DNA extraction kit for DNA extraction,the extraction of DNA PCR gene sequencing,use common fluorescent probe method is used to detect the SNP,design PCR primers for amplification including SNPS loci fragments and adjacent to the extension of SNP loci Primer,with online Primer Premier software design primers,Three loci of rs429358,rs7412 and rs7259620 were detected and typed,and the results were statistically analyzed.Results: A total of 200 patients with acute ischemic stroke and dyslipidemia who met the diagnostic criteria of traditional Chinese and Western medicine were screened out,including 133 males and 67 females,ranging in age from 32 to 90 years old,and the age distribution accorded with the normal distribution.All enrolled patients were dialectically dialectical,and the dialectical results were mainly phlegm and dampness,accounting for 70% of the total number.Blood stasis was the second,accounting for 52%.Internal fire and internal wind accounted for 33% and21%,respectively.Qi deficiency accounted for 35% and Yin deficiency accounted for 13%.Two alleles were detected at rs429358 and rs7412 in the coding region of Apo E gene.There were TT and CT alleles at rs429358[C/T]site.The TT frequency(82%VS.80%)and CT frequency(9%VS.4%)in the case group VS.the control group were detected.RS7412 [C/T] site has two alleles,CC and CT,CC frequency(82%VS.90%)and CT frequency(9%VS.2%)in case group VS.control group;There were three alleles at rs7259620[A/G] locus in the promoter region,which were GG,AG and AA,respectively.The frequency of GG(46%VS.65%),AG(50%VS.30%)and AA(4%VS.5%)in the case group VS.the control group.There was no significant difference in allele frequencies of the three SNP loci between the case group and the control group(P>0.05),and there was no significant correlation between the three SNP loci and the case group(P>0.05).Coding regions rs429358 and rs7412 two SNP loci polymorphism a total of four kinds of genetic phenotypes,cases of ε3ε3 respectively,ε2ε3,ε2ε4,ε3ε4 four phenotype,the control group with ε3ε3,ε2ε3,ε3ε4 three phenotypes,case group VS.control group of ε3ε3 genotype frequency(66%VS.70%),ε2ε3 genotype frequency(16%VS.10%),ε3ε4 genotype frequency(16%VS.20%),ε2ε4 genotype frequency(2%VS.0%),There was no significant difference in genotype distribution frequency between the two groups(P>0.05),and there was no significant correlation between genotype and case group(P>0.05).The ε3 allele frequency(82%VS.84%),the ε2 allele frequency(9%VS.5%)and the ε4 allele frequency(9%VS.10%)were not significantly different between the two groups(P>0.05),and there was no significant correlation between the genotype and the case group(P>0.05).In the correlation analysis of the promoter locus and coding locus,there were significant differences in the coding locus rs429358 among alleles of promoter locus rs7259620,but there was no significant correlation.However,the coding region rs7412 between alleles at rs7259620 in the promoter region was significantly different and correlated.Apo E genotyping frequency was significantly different among alleles at rs7259620 in the promoter region,and there was a significant correlation.In the correlation analysis of the three SNP loci and blood lipid,there was no significant difference between alleles of rs7259620 in the promoter region and no significant correlation between the blood lipid indexes.There were significant differences in the level of Apo A1 among alleles at rs429358 in coding region,and there was significant correlation.There were significant differences in the levels of HDL and Apo A1 between the two alleles at rs7412,and there was a significant correlation.There was a significant difference in the level of Apo A1 among all genotypes,and there was a significant correlation.Conclusion:(1)The syndrome differentiation of AIS patients with dyslipidemia is in line with the understanding of AIS patients with dyslipidemia based on the theory of traditional Chinese medicine.The main syndrome elements are phlegm and dampness syndrome,followed by blood stasis.(2)There was no correlation between rs429358,rs7412 and rs7259620 SNPs of Apo E gene and dyslipidemia of AIS with phlegm-dampness syndrome;(3)The polymorphism of rs7259620 in the promoter region can affect the expression of genotyping by affecting the polymorphism of rs7412 in the coding region;(4)TT gene carriers at rs429358 have higher levels of Apo A1,and CT gene carriers at rs7412 have higher levels of HDL and Apo A1,which have a protective effect on AIS.It can also be concluded that the ε2ε3genotype composed by the two have a protective effect on AIS.
