Diabetic Nephropathy(DN)is a common complication of diabetes and the main cause of end-stage renal disease.Sodium Butyrate(NaB)is a short-chain fatty acid,a metabolite of intestinal bacteria,and its protective effect on the kidneys in the case of DN has been widely reported.The purpose of this study is to study the effect of NaB on the whole transcriptome changes in DN and on DN.In our study,8-week male db/db mice with DN were randomly divided into two groups: DN +NaB group(DN mice treated with NaB at a dose of 5 g/kg/day)and DN group(DN mice)treated with saline).In addition,normal db/m mice were used as normal control(Normal control,NC).The blood glucose,body weight,urine microalbumin and urine creatinine of the mice were measured for all three groups.Full transcriptome analysis is performed by RNA Sequencing(RNA-Seq)to assess the profile of lnc RNA and mRNA.Bioinformatics analysis to predict potential lnc RNA and genes related to NaB in DN.Quantitative Real-time polymerase chain reactions(qRT-PCR)were used to detect the expression of lnc RNA and mRNA in kidney tissues and glomerular mesangial cells treated with NaB.The results of this study showed that NaB changed the expression of lnc RNA and mRNA in the kidney of DN mice,reduced the ratio of urine microalbumin to creatinine in DN mice,and improved the volume and volume of glomeruli in the kidney tissue of DN mice.Pathological processes such as thickening of the basement membrane.In addition,RNA-Seq results showed that,compared with the DN group,some lnc RNA and mRNA in the DN + NaB group had reverse changes.The co-expression network of lnc RNAs related to NaB revealed the interaction of these lnc RNAs with 155 key mRNAs.Through GO and KEGG analysis,it is found that most of these lnc RNAs and mRNAs are involved in the related processes of regulating inflammation.In addition,by further co-expressing the network of lnc RNA and mRNA constructs related to inflammation,the results show that lnc RNA Gm31284,Gm31278 and mRNA Psme1,Psme2 and Psme2 b play an important role in the inflammation response.All in all,this study shows that NaB can improve renal insufficiency caused by diabetes and regulate the transcriptome changes of DN.
