| Objective:Application in surface trauma hemorrhage rats focal different hemostatic materials(gelatin sponge,renewable oxycellulose(fiber silk)bleeding,by enzyme-linked immunosorbent experiment method for determining rat tissue inflammation factor,interleukin 6(IL 6)and the concentration of interleukin 2(IL-2),compare different kinds hemostatic materials affect tissue inflammation changes in rats.Methods:1.Establishment of rat brain model: 48 male SD rats,weighing300 ~ 350 g,were randomly divided into 3 groups(16 rats in each group):(1)Regenerated oxidized cellulose group,hemostasis was compressed with hemostasis gauze during operation.(2)Gelatin sponge group,intraoperative use of gelatin sponge compression hemostasis.(3)In the control group,intraoperative hemostatic materials were not used,only cotton pads were used for compression hemostasis.Each group was divided into 4 subgroups,with 4rats in each subgroup.They were sacrificed and detected at 7,14,21 and 28 days postoperatively,respectively.Rats with 10% chloral hydrate anesthesia after abdominal cavity,longitudinal cut skin,craniotomy in the right parietal bone(from the sagittal suture,coronary seam 5 mm)with dental drill grinding carefully polished out a 5 mm * 3 mm size of bone window,full hard membrane is not damaged,open the epidural,don’t touch the cortex,the brain tissue exposed and epidural hemorrhage with wet cotton piece of oppression hemostasis.Microscopic tumor forceps were used to clamp the cerebral cortex in a range of 2mm×2mm×1mm,causing brain tissue trauma and bleeding.According to the groups,cotton pad compression,gelatin sponge and regenerated oxidized cellulose were applied to stop the bleeding,and the wound was sutured after complete hemostosis.The rats were given intraperitoneal injection of penicillin to prevent infection on day 1,2 and 3 after surgery,and the rats were fed normally after surgery.Four rats in each group were sacrificed on day 7,14,21 and 28,respectively,and the brain tissue samples were taken for the next experiment.1.The levels of interleukin 6(IL-6)and interleukin 2(IL-2)in rat brain tissues were detected by enzyme-linked immunosorbent assay,and the degree of inflammatory response was quantified and compared by measuring absorbance value.Results:1.In the gelatin sponge group,the concentrations of interleukin 6(IL-6)and interleukin 2(IL-2)increased from 7 days(1463.31±49.19;997.84±139.85)and reached the peak value(3565.37±56.08 on the 14 th day;1463.34±58.75)and then decreased gradually(1089.36±202.07;794.46 ± 46.82).2.In the control group,the concentrations of interleukin 6(IL-6)and interleukin 2(IL-2)increased from 7 days(738.53±226.92;774.16±85.27),and reached the peak value on the 21 st day(3546.15±28.98;1059.28±33.19)and then decreased gradually(1491.86±220.51;869.58 ±84.77).3.In the regenerated oxidized cellulose group,the concentrations of interleukin 6(IL-6)and interleukin 2(IL-2)increased from day 7(810.10±211.02;869.58±84.77)began to increase,and reached the peak value on the 21 st day(3304.61±306.06;1164.38±167.69)and then decreased gradually(848.99±328.46;869.58 ± 84.77).4.Comparison of interleukin-6(IL-6)concentration between the regenerated oxidized cellulose group and the control group showed P > 0.05,which was not statistically significant.Regenerated oxidized cellulose did not cause the change of interleukin-6(IL-6)in the traumatic brain tissue.5.Comparison of interleukin 6(IL-6)concentration between the gelatin sponge group and the control group,P < 0.05,was statistically significant;Gelatin sponge can cause the increase of interleukin-6(IL-6)in traumatic brain tissue.6.Comparison of interleukin 2(IL-2)concentrations in regenerated oxidized cellulose group,gelatin sponge group and control group,P < 0.05,was statistically significant;Gelatin sponge can cause the increase of interleukin-2(IL-2)in traumatic brain tissue.Conclusion:Interleukin-6(IL-6)and interleukin-2(IL-2)were not altered by regenerated cellulose after traumatic brain hemorrhage in rats.However,interleukin-6 and interleukin-2 were increased by gelatin sponge. |
