| The ocean covers 71%of the earth and is the cradle of life.At the same time,80%of the earth’s total biomass come from the ocean,which is about 30 phylums incluing more than 2.5 million species.The ocean has a special environment,due to hypoxia and dark environment,marine organisms formed unusual evolution pathways and enzyme reaction systems,which are quite different from terrestrial organisms-As for this we can speculate that the marine life should contain better specific secondary metabolites which are different from land-based biological origin,having a novel chemical structure and significant biological activity.Bio film-mediated drug resistance is a major cause of chronic infection and clinical recurrence.The study on the mechanism of bio film resistance at home and abroad is still mature,which is based on its resistance mechanism and the structure to find ways to destroy the biofilm,resulting in a research point about biofilm related infections.As an important source of marine natural products,we have found a large number of active compounds in gorgonian,including steroids,diterpene,sesquiterpene,prostaglandins,alkaloids and so on.Exploring the metabolites,damaging effects of bio films on studying new drugs is very important.The currenr study on the chemical investigation of the gorgonian Junceella fragilis which is from the South China Sea leaded to secondary metabolites.We can use the research methods and modern technology spectrum on marine natural products chemistry-on preparing for separation,structure identification and chemical analysis.Driven by DAD-HPLC and 1H-NMR,a more systematic chemical investigation of gorgonian coral Junceella fragilis aided by normal phase chromatography on silica gel column,ODS column chromatography and semipreparative high performance liquid chromatography methods were conducted.And by using two dimensional NMR correlation spectroscopy(HSQC,HMBC,COSY)and other spectroscopic spectra.Binding the effection of biofilm to be a further comprehensive analysis,designed to provide a basis for drug development Fifteen compounds were insolated from the gorgonian coral Junceella fragilis using modern chromatography,mainly terpenes.Among them,compound 1-6 and compound 13 are briarane diterpene,compound 7 is terrein,compound 8-9 are Iso-coumarin,Iso-coumarin 10-12 are sesquiterpenes and compound 14-15 are cembranoid diterpenes.To investigate whether the isolated compounds exhibit activity on the biomembrane,five strains including Pa,Ec,Ca,and Sa were selected and were cultivated at 37℃.The fifteen compounds isolated from the gorgonacea Junceella fragilis was added to the strains.The effects caused by the compounds displayed very weak effect. |
