| Objective The purpose of this study was to improvement of masticatory muscles during isometric,isotonic contraction of animal model and establishment of standard process of masseter muscle fibers and mitochondria,the isometric,isotonic contraction force adaptive remodeling.Methods 1.The improved model the animal path and method,according to the different experimental animal to individual can standard unified modeling requirements 2.The experimental object worn device after 8 weeks,the muscle fiber material,morphological changes were observed under light microscope by HE staining 3.After 8 weeks,the immunohistochemical staining of actin,myosin staining,comparative analysis of gray value.4.After 8 weeks,the transmission electron microscope magnified 1000 times for each experimental group mitochondria count,transmission electron microscope to observe the morphological changes of mitochondria enlarged 3000 times.Results 1.We Successfully improved the animal experimental model.2.HE staining indicated that the experimental group compared with the control group,masseter muscle fibers became thicker,the isotonic contraction muscle fiber bundles thickening was more obvious 3.The statistical results show that the isometric group,isotonic contraction of actin group compared with the control group did not change significantly,and there is a significant change of myosin.4.The results showed that the experimental group intermyofibrillar mitochondria decreased compared with the control group,with isotonic contraction group decreased more significantly;mitochondria isometric,isotonic differences between the three groups was more than that in control group,the isometric group was more obvious.Conclusions 1.The improved and standardized animal models of masticatory muscle isometric and isotonic contraction can meet the demand of the research.2.After Isometric,isotonic contraction exercise,masseter muscle fibers were thickened,with isotonic contraction group was more significant.3.After masseter muscle Isotonic and isometric contraction exercise,compared with the control group,the experimental group did not change significantly and actin,myosin appear obvious change.The specific reasons to be further molecular biology research to support.4.After Isometric,isotonic contraction exercise,muscle mitochondria also motion adaptive remodeling.Transmission electron microscope magnified 30000 times field observed between the myofibrils,mitochondria morphology than that in the control group were changed;at the same time,transmission electron microscope magnified 10000 times from the perspective of counting,the statistical results show that the experimental group intermyofibrillar mitochondria decreased compared with the control group,with a shrinking group decreased more significantly;mitochondria length,etc.Zhang differences between the three groups was more than that in control group,the isometric group was more obvious.5.There are difference in alteration between masticatory muscles after isometric and isotonic contraction。A large number of satellite cell proliferation and differentiation,in isometric exercise induced muscle cells in the proliferation,and in isotonic contraction exercise induced muscle cells mainly to fusion. |
