Abstract/Summary:
Background and objective The liver cancer is kind of a worldwide malignant tumor which has a fifth prevalence rate,a second mortality rate,high degree of malignancy and poor prognosis among all kinds of cancers.IARC published data show that there are nearly 1 million cases of newly diagnosed patients with liver cancer,about 690 thousands people died of liver cancer in the world every year.What’s more the liver cancer incidence rate continues to rise,causing a great threat to global human health.In China,the incidence of liver cancer ranks fourth and the death’s rate ranks second among kinds of malignant tumors.At present,many traditional Chinese medicine clinical practice shows that a variety of traditional Chinese medicine and there extracts in the treatment of malignant tumors have quite good results.There are 234 commonly used prescription for clinical treatment of liver cancer,containing 280 kinds of common medicine,which use peony 73 times.Studies have confirmed that the extract of peony has a variety of biological activities,including tumor inhibition,but there is no report on the anti-tumor activity of the albiflorin.In this study,using oxaliplatin as positive control drug,the study try to investigate the effects of albiflorin on proliferation and apoptosis of SMMC7721 cells in hepatocellular carcinoma.Methods and materials Divide the SMMC-7721 cells into the following groups:1)the albiflorin group,further divided into low(0.25g/L),middle(0.5g/L)and high(1g/L)dose;2)the oxaliplatin group,32μM/L;3)the combination group,further divided into low(32μM/L oxaliplatin+0.25g/L albiflorin),middle(32μM/L oxaliplatin+0.5g/L albiflorin)and high(32μM/L oxaliplatin+1g/L albiflorin)dose;4)the control group.The HE staining was sued to observed the morphological change of the cells,MTT assay was used to determine the inhibition under 24h,48h,72h,respectively.The flow cytometry and DNA Ladder were used to analyze the apoptosis.Results HE staining showed that after 48h of administration,the morphological change of the cells was obvious.MTT assay showed that albiflorin can apparently inhibit the proliferation of the cells under different doses,and the effect was time-dose depended.The flow cytometry and DNA Ladder both showed the apoptosis induced by albiflorin was obvious.The oxaliplatin mainly inhibited the S stage of the SMMC7721 cells,while albiflorin mainly inhibited the G2/M stage,the combination group inhibited both S and G2/M stage.Conclusion In conclusion,albiflorin shows a good effect for inhibiting the proliferation and inducing the apoptosis of SMMC7721 cells in hepatocellular carcinoma.
