| Objective:To study the effect of compound extract on Bax/ Caspase-3/Fas/Fas L in the RCS rat with syndromes of blood stasis and deficiency and to explore its mechanism,to provide more evidence for the modern medicine in the treatment of retinitis pigmentosa.Methods:24 RCS rats were divided into normal group,model group,Ziyinmingmu group(8 each).Normal group: female RCS(rdy+/+,p+/+)rats(n=4),male RCSrdy+/+,p+/+)rats(n=4),filling the stomach with 0.9%NS.Model group: female RCS(rdy-/,p-/-)rats(n=4),male RCS(rdy-/-,p-/-)rats(n=4),filling the stomach with 0.9%NS.Ziyinmingmu group: female RCS(rdy-/-,p-/-)rats(n=4),male RCS(rdy-/,p-/-)rats(n=4),filling the stomach with Ziyinmingmu Pill.After filling the stomach 30 days,detect the expression of Bax/ Caspase-3/Fas/Fas L in the retinal tissue by immunohistochemical method,detect retinal tissue Bax/ Caspase-3/Fas/Fas L proteins relative content by Western blot,determinate the expression of Bax/Caspase-3/Fas/Fas Lm RNA by Real-time q PCR.Results : The slice of model group retinal pathology organization dyed by HE showed that there are disorder of retinal outer structure.The slice of retinal pathology organization dyed by HE of the RCS(rdy-/,p-/-)rats in Ziyinmingmu group showed that there are improvement on disorder of retinal outer structure(P <0.05).Compared with the normal group,model group rats Bax/ Caspase-3/Fas/Fas L m RNA expression increased significantly(P < 0.01).Compared with model group,Ziyinmingmu group can significantly reduce the expression levels of Bax/Caspase-3/Fas/Fas L m RNA(P < 0.01).Compared with the normal group,model group rats Bax protein and Caspase 3 protein expression also increased significantly(P < 0.01).Compared with model group,Ziyinmingmu group can significantly reduce the expression levels of Bax protein,Caspase 3 protein,Fas protein and Fas L protein(P<0.01).Conclusion: The following conclusions were drawn through the experimental research:Ziyinmingmu Pill can significantly reduce the cell apoptosis in the retinal tissue. |
