| Background: Nasopharyngeal carcinoma(NPC)is caused by a combination of factors: viral,environmental influences,and heredity.The viral influence is associated with infection with Epstein-Barr virus(EBV).The Epstein-Barr virus is one of the most common viruses.95 percent of all people in the U.S.are exposed to this virus by the time they are 30–40 years old.The World Health Organization does not have set preventative measures for this virus because it is so easily spread and is worldwide.Very rarely does Epstein-Barr virus lead to cancer,which suggests a variety of influencing factors..Other likely etiological factors include genetic susceptibility,consumption of food(in particular salted fish)containing carcinogenic volatile nitrosamines.Objective:(1)This study was aimed q PCR detection of differentially expressed paraffin nasopharyngeal nasopharyngeal mucosa with chronic inflammation paraffin mi R-19 b and mi R-19 b so as to explore the pathogenesis of NPC.(2)Immunohistochemical detection of nasopharyngeal specimens,and nasopharynx mi R-19 b target gene product in the mucosa of chronic inflammation paraffin SOCS-1 differentially expressed mi R-19 b and thus the molecular pathways for scientific research.Methods:(1)taking nasopharyngeal mucosa specimens of patients with chronic inflammatory paraffin,paraffin 60 nasopharyngeal carcinoma.Then total RNA extraction,reverse transcription,mi R-19 b of q PCR,ssps analyze the expression of mi R-19 b paraffin difference between the mucosa of patients with chronic inflammation of the nasopharynx,nasopharyngeal carcinoma patients.(2)take nasopharyngeal mucosa specimens of patients with chronic inflammatory paraffin,paraffin blocks of nasopharyngeal carcinoma by immunohistochemical detection difference SOCS-1 expression and correlation analysis and differential expression of mi R-19 b paraffin explore mi R-19 b in nasopharyngeal carcinogenesis in molecular pathways.Results:(1)on the expression of mi R-19 b nasopharyngeal paraffin into the q PCR detected,compared with nasopharyngeal mucosa of chronic inflammation paraffin,mi R-19 b expression abnormalities(P <0.05),and prompt mi R-19 b overexpression.(2)Immunohistochemical detection of patients with chronic inflammatory nasopharyngeal mucosa of patients with nasopharyngeal carcinoma paraffin show,SOCS-1 expression in NPC were low,but there are patients with high expression in the mucosa of patients with chronic inflammation,and mi R-19 b in q PCR detection of consistency,indicating that differential expression of mi R-19 b may be associated with SOCS-1,statistics show that nasopharyngeal paraffin tissue blocks SOCS-1 mi R-19 b and the correlation coefficient is negative correlation.Conclusions:(1)comparison of the mucosa of patients with chronic inflammation of the nasopharynx paraffin,mi R-19 b overexpressed in nasopharyngeal carcinoma patients,statistically significant(P <0.05).So paraffin in mi R-19 b promising as a molecular marker of nasopharyngeal carcinoma clinical diagnosis.(2)negative correlation of mi R-19 b and SOCS-1 instructions,may explain mi R-19 b by degradation of the target gene,resulting in target gene product SOCS-1 translation is blocked,thereby affecting the STAT pathway on proliferation and apoptosis of cells. |
